摘要
目的:筛选益肾化瘀方拮抗肾间质纤维化作用的有效部位。方法:将益肾化瘀方煎煮液采用不同极性的有机溶剂进行萃取分离,分别得到益肾化瘀方石油醚部位、益肾化瘀方乙酸乙酯部位、益肾化瘀方正丁醇部位和水饱和部位。同时将大鼠随机分为假手术组、模型组、益肾化瘀方不同提取部位组、缬沙坦组。除假手术组外,其余6组复制单侧输尿管梗阻模型,比较各给药治疗组在不同时间点对大鼠肾功能、肾脏病理及肾组织中羟脯氨酸含量的影响。结果:大鼠梗阻后均出现明显的肾小管萎缩和肾间质纤维化,并伴有血清尿素氮、血肌酐、胱抑素C水平的升高,且随梗阻时间的延长而逐渐加重。给予益肾化瘀方4个不同极性部位提取物及缬沙坦治疗后,大鼠肾间质细胞炎性浸润、肾小管萎缩和肾间质纤维化病变逐渐减轻,肾功能明显改善,肾组织中羟脯氨酸含量降低。而在改善大鼠肾脏病理损伤、改善肾功能、降低肾组织羟脯氨酸含量方面益肾化瘀方正丁醇部位组疗效与缬沙坦组相当,显著优于其余3个极性部位。结论:益肾化瘀方正丁醇部位为本方发挥抗肾间质纤维化作用的有效部位。
Objective: To screen effective parts on antagonism of Yishen Huayu Fang to renal interstitial fibrosis( RIF). Methods:The water solution of Yishen Huayu Fang was extracted and separated by different polarity organic solvent,and petroleum ether polar area,ethyl acetate polar area,n-butanol polar area and water saturation polar area of Yishen Huayu Fang was identified respectively. Rats were randomly divided into control group,model group,valsartan group and Yishen Huayu Fang different extraction parts treatment group. Except the control group,the other 6 groups rats were established unilateral ureteral obstruction( UUO)model. The kidney function,renal pathology and the level of hydroxyproline in kidney tissues at different time points of all groups were compared. Results: All rats appear obvious renal tubular atrophy and renal interstitial fibrosis after UUO established,and accompanied by the rise of BUN,Scr and Cys C level,and levels were higher when prolonged obstruction. Renal interstitial inflammatory cell infiltration,renal tubular atrophy and renal interstitial fibrosis pathological changes gradually reduced,renal function improved significantly and the level of hydroxyproline in kidney tissues reduced after four different polar parts extraction of Yishen Huayu Fang and valsartan were treated. Rat renal pathological tissue damage,renal function and renal tissue hydroxyproline level were all improved in both n-butanol part group of Yishen Huayu Fang and valsartan group,and significantly better than the level of the rest three polar parts. Conclusion: N-butanol part is a effective part of Yishen Huayu Fang on antagonism to renal interstitial fibrosis.
