摘要
目的:检测三七总皂苷对人脐静脉内皮细胞损伤后的影响,探索三七总皂苷对内皮细胞的保护作用。方法:把体外培养的人脐静脉内皮细胞株传代培养,共分为6组,即正常组,模型组,三七总皂苷4个浓度组,用10μg·L^(-1)的肿瘤坏死因子-α(tumor necrosis factor,TNF-α)造成细胞损伤模型,三七总皂苷组分别加入不同浓度的三七总皂苷2.5 mg·L^(-1)、5 mg·L^(-1)、10 mg·L^(-1)、20 mg·L^(-1),用CCK-8方法检测各组细胞活性。结果:CCK-8检测结果显示,人脐静脉内皮细胞在TNF-α诱导损伤后,用4个浓度的三七总皂苷干预,随着时间的延长,细胞存活率逐渐降低,组间差异具有统计学意义,其中6 h的细胞存活率最高,24 h的细胞活性最差,差异无统计学意义;和模型组相比,三七总皂苷4个浓度组细胞活性明显较高(P<0.01或P<0.05),其中5 mg·L^(-1)三七总皂苷组细胞活性最高,与模型组相比,差异具有统计学意义(P<0.01)。结论:三七总皂苷能提高人脐静脉内皮细胞损伤后的活性。
Objective: Detection of notoginseng total saponin on human umbilical vein endothelial cells after injury and the effect of exploration notoginseng total saponin on protection of endothelial cells. Methods: The in vitro cultured human umbilical vein endothelial cell line subculture,which is divided into six groups,namely the normal group,model group,notoginseng total saponin concentrations of four groups,with 10 mg·L-1of tumor necrosis factor alpha( TNF-α) cells damage model,notoginseng total saponin group respectively in different concentrations of total saponins of panax notoginseng 2. 5 mg·L-1,5 mg·L-1,10 mg·L-1,20mg·L-1,CCK-8 method was used to inspect each cell activity. Results: CCK-8 test results showed that human umbilical vein endothelial cell injury induced by TNF-α,with notoginseng total saponin of four concentration,with the extension of time,the survival rate of cells decreased gradually,with a significant difference between the groups,of which 6 h cell survival rate was the highest,24 h cell activity difference was the worst,with no statistically significant; compared with the model group,group activity of Notoginseng Total Saponin 4 concentration was significantly higher( P 〈0. 01 or P 〈0. 05),the total saponins of 5 mg·L-1concentration group were the highest activity,compared with the model group,the differences were statistically significant( P 〈0. 01).Conclusion: Panax notoginseng total saponin can improve the activity of human umbilical vein endothelial cell after injury.
