摘要
目的探寻连翘叶中指标性成分,建立指标性成分的含量测定方法,并对比连翘叶不同提取物中连翘酯苷A的含量。方法采用高效液相色谱法测定连翘酯苷A的含量。色谱柱为十八烷基硅烷键合硅胶,以乙腈-0.4%醋酸水(18∶82)溶液为流动相,检测波长为330 nm,柱温25℃。结果连翘酯苷A进样量在1.047~20.94μg线性关系良好(r=0.9998);平均加样回收率为104.10%(RSD 2.36%)。连翘叶70%乙醇、甲醇、水三种不同提取物中连翘酯苷A的含量分别为5.36%、8.16%、3.32%。结论该方法准确、可靠,可用于连翘叶药材及相关产品中连翘酯苷A的含量测定,为完善连翘叶药材质量标准提供参考。
Objective To explore the ingredient index of forsythia suspense leaves,establish a method for determination of the index component,and compare the content of forsythoside A in different extracts of Forsythia leaves. Methods High performance liquid chromatography was used to determine the content of forsythoside A. The chromatographic column was eighteen alkyl silane bonded silica gel,and acetonitrile-0. 4% acetic acid water( 18 ∶ 82) was used as mobile phase. The detection wavelength was 330 nm and the column temperature was 25℃. Results Forsythiaside A sample volume has linear relationship between 1. 047 and 20. 94 μg( r =0. 999 8),the average recovery rate was 104. 10%( RSD 2. 36%). The content of forsythoside A in different extracts( 70% ethanol,methanol and water) of forsythia leaves were 5. 36%,8. 16% and 3. 32% respectively. Conclusion The method is accurate,reliable and can be used for the determination of forsythoside A in the leaves of forsythia and related products,which provides a reference for improving the quality standard of forsythia leaves.
