摘要
为进一步研究CPT2基因在脂质代谢中的作用,试验构建了绿色荧光CPT2过表达载体,将其转入奶牛乳腺上皮细胞,通过荧光定量方法,检测CPT2基因mRNA的表达水平差异,在细胞水平验证了CPT2过表达载体的表达效率。结果表明:过表达组的CPT2基因mRNA表达量与对照组(空载组)相比有所提高。说明CPT2过表达载体转染奶牛乳腺上皮细胞后,CPT2基因mRNA表达水平显著升高。
The aim of the present study was to further investigate the role of CPT2 gene in lipid metabolism. The green fluorescence Camitine Palmitoyhransferase 2 (CPT2) overexpression plasmid was constructed and transfected into the mammary epithelial cells (MEC). The mRNA expression level of CPT2 gene was detected by fluorescence quantitative PCR method. The expression efficiency of CPT2 overexpressing vector was verified at the cellular level The results showed that the expression of CPT2 mRNA in the overexpression group was sigrtificantly higher than that in control group. The results indicated that the mRNA expression level of CPT2 gene was significantly increased after transfection of CPT2 overexpression vector in bovine mammary epithelial ceils.
