摘要
为了研究miRNA表达量与肉质风味表型指标的相关性,试验采用实时荧光定量PCR(qRTPCR)检测了藏猪和大约克夏猪背最长肌中miR-27a、miR-378的差异表达。结果表明:藏猪在6月龄时背最长肌中miR-27a和miR-378的表达量均极显著低于大约克夏猪(P<0.01)。相关性分析显示,miR-27a、miR-378与肌内脂肪含量和硫胺素呈负相关,而与肌纤维直径、眼肌面积和瘦肉率呈显著或极显著正相关(P<0.05或P<0.01)。说明miR-27a和miR-378对猪肉品质具有重要的负调控作用。
To study the correlation between miRNA expression level and phenotypic indexes of meat quality ,quantitative real - time PCR( qRT - PCR ) method was adopted in this experiment in determining differential expression of miR -27a and miR -378 in longissimus dorsi between Ti- betan pigs and Large Yorkshire pigs. The results showed that miR - 27a or miR - 378 expression levels in longissimus dorsi at 6 months old were very significantly lower from Tibetan pigs than those from Large Yorkshire pigs( P 〈 0.01 ). Correlation analysis showed that miR -27a or miR - 378 expression levels were in negative relation with intramuscular fat contents or thiamine, while were in significant or very significant pos- itive relation with muscle fiber diameter,eye muscle area or lean meat rate( P 〈 0.05 or P 〈 0.01 ). The results suggested that miR -27a or miR- 378 expression had an important role in negative regulation on meat quality.
