獭兔FSHβ基因的多态性及其与繁殖性状的关联性研究 被引量:4

摘要 为了研究卵泡刺激素(FSHβ)基因与獭兔繁殖性状间的关系,将其作为獭兔繁殖性状的候选基因进行多态性检测与关联分析,试验以100只海狸色獭兔和70只白色獭兔为试验材料,采用PCR扩增测序以及Snapshot分型技术对獭兔FSHβ基因进行遗传多态性分析,通过PCR测序法鉴定了FSHβ基因第1外显子区、第3外显子区内的单核苷酸多态性(SNPs)位点,用最小二乘法建立分析模型,利用SPSS 17.0软件分析数据。结果表明:在170只个体中,共筛选出5个SNPs,其中有2个位于1号外显子(FSH1、FSH2),3个位于3号外显子(FSH25、FSH30、FSH31)。FSH25与初产仔数、产活仔数关联分析显著(P<0.05),FSH31与产活仔数、初产窝重、21日龄活仔数关联分析显著(P<0.05)。这2个位点可以作为分子标记对獭兔进行标记辅助选择。 In order to study the relationship between FSHβ gene and reproductive traits in Rex rabbit ,and to make this gene as a candidate in palymorphism investigation and correlation analysis of reproductive traits in Rex rabbit, 100 beaver Rex rabbits and 70 white Rex rabbits was used in this experiment. PCR amplification sequencing and Snapshot technology methods were carried out in analyze genetic polymorphism of FSHβ gene of Rex rabbits. PCR amplification sequencing identified single nucleotide polymorphisms(SNPs) in Exon 1 and Exon 3 of FSHβ gene with least square method for model building and SPSS 17.0 software for data analysis. The results showed that 5 SNPs were obtained from 170 individuals,in which 2 of them was located in Exon 1 ( FSH1, FSH2 ) , and the other 3 SNPs in Exon 3 ( FSH25, FSH30, FSH31 ). Correlation analysis showed that FSH25 was significantly relative with new born number post delivery or live new born number(P 〈 0.05 ) ;FSH31 was in significant relation with live new born number,litter weight or 21 -day- old live new born number(P 〈0.05). Those 2 SNPs could be selected as genetic makers in marker assisted selection of Rex rabbit.
