内蒙古呼伦贝尔地区优势蜱种的鉴定及其基因多态性分析 被引量:4

Identification and gene polymorphism analysis of dominant ticks in Hulun Buir,Inner Mongolia

摘要 为掌握内蒙古呼伦贝尔地区硬蜱的优势蜱种类,对该地区15个调查点的蜱进行考查和采集蜱虫1 349只样本,在形态学鉴定方法初步进行分类的基础上,设计1对简并引物,从蜱虫基因组DNA样品中,采用PCR扩增得到蜱虫16SrRNA基因序列,测序后进行基因序列同源性分析。用Mega 6.06软件绘出系统进化树,与GenBank数据库中已知蜱虫16SrRNA进行聚类。结果显示:15个考察点蜱虫种类均为草原革蜱,属于呼伦贝尔地区常见优势蜱虫之一。但是,不同地区的蜱虫之间也存在基因序列的差异性。 In order to master advantage speeie of ixodes in Hulun Buir,Inner Mongolia,where 1 349 samples from 15 survey spots were identifiled and collected,a pair of degenerate primer from DNA of tick's samples was designed after classified in morphology level and then obtained gene sequences of tick's 16S rRNA by PCR method, and subsequently sequenced phylogenetic tree was constructed by Mega 6.06 and clustered by their respective 16S rRNA in GenBank. The results revealed that all tick species in 15 research areas were Dermacentor nuttalli and it was one of the most common ticks in Hulun Buir. But there is also a different gene sequence between ticks from different areas.
