猪圆环病毒2型LAMP检测方法的建立和初步应用

摘要 根据GenBank上已报道的猪圆环病毒2型(PCV2)基因组序列设计1组LAMP引物(F3/B3、FIP/BIP),通过对环介导等温扩增技术(LAMP)反应条件的优化,建立了猪圆环病毒2型LAMP检测方法,并对阳性结果进行了酶切验证。结果表明:该方法具有较高的检测灵敏度,最低可以检测到反应体系内1 pg的PCV2DNA;通过反应结束后的肉眼可视化观察,可方便地进行结果判定。本研究建立的PCV2 LAMP检测方法能够满足基层临床PCV2病原的快速检测要求。 A group of LAMP primers(F3/B3,FIP/BIP) were designed according to the genome sequences of porcine circovirus type 2(PCV2) reported on GenBank. The LAMP method for detecting porcine circovirus type 2 was established by optimizing the reaction conditions of loop mediated isothermal amplification(LAMP),and the positive results were verified by enzyme digestion. The results showed that the method had high detection sensitivity, and the PCV2 DNA of 1 pg in the reaction system could be detected at minimum. After the end of the reaction,the visual observation of naked eye could be used to judge the result conveniently. The PCV2 LAMP method established in the study could meet the requirements of rapid clinical detection of PCV2 pathogens.
