红花械离体培养及标准化繁殖

摘要 以带腋芽的茎段为外植体,对2个红花槭良种进行离体培养研究。结果表明:初代培养基MS+2ip0.5 mg/L+GA3 0.5 mg/L较适宜;最佳增殖培养基为WPM+2ip 1.0 mg/L+TDZ 0.002 mg/L+GA_3 0.5 mg/L,最佳培养温度为28℃,在此条件下,2个红花槭品种的增殖率分别为1.95和1.90;生根培养基WPM+IBA 0.5mg/L+NAA 0.5 mg/L效果较佳,2个红花槭品种的生根率分别达85%和90%;适时移栽成活率可达80%以上。 Taking the stem segments with axillary buds as explants, 2 fine cultivars of Acer rubrum was studied in vitro. The results showed that primary culture medium MS +2 ip 0. 5 mg/L + GA3 0. 5 mg/L was more appropriate, the best multiplication medium was WPM + 2 ip 1.0 mg/L + TDZ 0. 002 mg/L + GA3 0. 5 mg/L, the optimum culture temperature was 28 ℃. Under these conditions, the coefficient of multiplication of two Acer rumrum cultivars were 1.95 and 1.90, respectively. The rooting medium WPM + IB A 0. 5 mg/L + NAA 0.5 mg/L was better,and the rooting rates of 2 Acer rumrum cultivars were 85% and 90%,respectively. The survival rate of timely transplanting could reach more than 80%.
