摘要
为铁皮石斛药效及多糖药理活性提供科学依据,采用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化方法使糖类物质带上紫外基团,用高效液相色谱仪紫外检测器进行检测,对贵州不同产地铁皮石斛单糖组成进行测定,并对其中单糖的含量进行分析。结果表明:铁皮石斛多糖主要由甘露糖、葡萄糖、半乳糖醛酸和半乳糖等组成,其中甘露糖和葡萄糖含量较高,甘露糖均占总多糖35%以上,葡萄糖均占总多糖12%以上,贵州不同产地铁皮石斛药材中各单糖占总多糖含量的比例存在明显差异,大方县产铁皮石斛单糖含量相对较低,而乌当及独山等地产铁皮石斛单糖含量相对较高。
The composition and content of monosaccharide in D.officinale from different producing areas in Guizhou were detected by the ultraviolet detector of HPLC after the carbohydrates are carried with ultraviolet radicals by PMP pre-column derivatization method to provide the scientific basis for pharmacological function and polysaccharide pharmacological activity of D.officinale.Results：The polysaccharide of D.officinaleis composed by mannose,glucose,galacturonic acid and galactose mainly.The content of mannose and glucose accounts for above 35%and 12% of total polysaccharide respectively.There is a significant difference in proportion of different monosaccharide in total polysaccharide of D.officinale from different producing areas.The monosaccharide content of D.officinale from Dafang County is lower relatively and monosaccharide content of D.officinale from Wudang District and Dushan County is higher.
