摘要
目的探讨促性腺激素释放激素α(GnRH-α)对子宫内膜异位症患者黄体中期子宫内膜细胞HOXA10表达的影响。方法选取2014年1月至2016年1月治疗的因子宫腺肌病或卵巢子宫内膜异位囊肿手术治疗的患者10例(观察组),已正常生育的非子宫内膜异位症、非子宫内膜病变、非恶性肿瘤的手术患者6例(对照组)。收集两组患者黄体中期子宫内膜标本,空白组处理后用GnRH-α进行预处理。比较两组子宫内膜间质细胞存活率和凋亡率;检测两组子宫内膜间质细胞HOXA10表达。结果观察组和对照组在同种处理条件下子宫内膜间质细胞存活率比较差异无统计学意义(P>0.05);GnRH-α对观察组和对照组子宫内膜间质细胞存活率无影响(P>0.05);观察组和对照组在同种处理条件下子宫内膜间质细胞凋亡率比较差异无统计学意义(P>0.05);GnRH-α处理下观察组和对照组子宫内膜间质细胞凋亡率分别为(0.041±0.013)%和(0.045±0.020)%,均明显低于空白对照(P<0.05);同种处理条件下的观察组和对照组子宫内膜间质细胞HOXA10表达△CT值比较差异有统计学意义(P<0.05);GnRH-α处理下观察组和对照组子宫内膜间质细胞HOXA10表达△CT值分别为(7.912±0.017)%和(5.510±0.019)%,均明显高于空白对照(P<0.05);GnRH-α处理下观察组子宫内膜间质细胞HOXA10表达△CT值明显高于对照组(P<0.05)。结论体外GnRH-α处理可以增加子宫内膜间质细胞HOXA10的表达,可能对子宫内膜容受性有一定改善。
Objective To investigate the effect of vitro pretreatment of GnRH-α on HOXA10 expression of midluteum endometrial stromal cell in endometriosis patients. Methods From January 2014 to January 2016,10 cases of patients with adenomyosis or ovarian endometriosis cyst in our hospital for treatment( observation group) and 6 patients with normal fertility and non endometriosis,non endometrial lesions and non malignant tumors( control group) were selected in this study. Mid-luteal endometrial specimens were collected,GnRH-α were used to act on endometrial cells,HOXA10 expression were detected. Results The survival rate of endometrial stromal cells of observation group and control group had no statistical significant difference under the same pretreatment( P 〉 0. 05). GnRH-α had no effect on the survival rate of endometrial stromal cells in the observation group and the control group( P 〉 0. 05). The apoptosis rate of endometrial stromal cells of observation group and control group had no statistical significant difference under the same pretreatment( P 〉 0. 05). Under the GnRH-α pretreatment,the apoptosis rate of endometrial stromal cells in the observation group and control group were( 0. 041 ± 0. 013) % and( 0. 045 ± 0. 020) %,which were significantly lower than that the blank control( P 〈 0. 05). Under the same pretreatment conditions,△CT value of HOXA10 expression of endometrial stromal cells in observation group and control group had statistical significant difference( P 〈 0. 05). Under the GnRH-α pretreatment,endometrial stromal cells HOXA10 expression △CT values in the observation group and the control group were( 7. 912 ± 0. 017) % and( 5. 510 ±0. 019) %,significantly higher than that of the blank control( P 〈 0. 05). Under GnRH-α pretreatment,the observation group endometrial stromal cells HOXA10 expression △CT value was significantly higher than that of control group( P 〈 0. 05). Conclusion Vitro GnRH-α pretreatment can increase the expression of HOXA10 in endometrial stromal cells,may improve the endometrial receptivity.
