摘要
目的:研究见血清中的化学成分并进行分离鉴定。方法:将见血清药材粉碎,用95%乙醇在85℃下回流提取,所得提取物减压浓缩回收溶剂得总浸膏,再将其用蒸馏水分散,然后依次用等体积石油醚、乙酸乙酯和正丁醇进行萃取,得到石油醚、乙酸乙酯和正丁醇萃取部位。运用硅胶柱色谱,LH-20羟丙基葡聚糖凝胶(Sephadex LH-20)柱色谱和半制备高效液相色谱对石油醚和乙酸乙酯萃取部位进行分离纯化,获得单体化合物。并根据单体化合物理化性质和波谱数据鉴定化合物的结构。结果:从见血清95%提取物的石油醚和乙酸乙酯萃取部位中分离得到12个化合物,分别是对羟基苯甲酸(1),原儿茶酸(2),对羟基肉桂酸(3),反式阿魏酸二十二烷酯(4),顺式阿魏酸二十二烷酯(5),豆甾-4-烯-3-酮(6),橙皮内酯水合物(7),花椒毒酚(8),橙皮油内酯烯(9),豆甾醇-3-O-β-D-吡喃葡萄糖苷(10),香草酸(11)和对羟基苯甲醛(12)。结论:化合物1~10为首次从羊耳蒜属植物中分离得到,化合物11为首次从脉羊耳兰中分离得到。
Objective: To study the chemical constituents of the whole plant of Liparis nervosa. Method:The herb L. nervosa was crushed into powders and then extracted with 95% Et OH at 85 ℃. After removal of Et OH under reduced pressure,the solvent was recovered to obtain total extract. The extract was then suspended in distilled water and partitioned successively with petroleum ether( PE),Et OAc,and n-Bu OH to get corresponding fractions. Silica gel column chromatography( CC),Sephadex LH-20 gel CC and semi-preparation HPLC were used for the separation and purification of PE and Et OAc fractions. The structures of the obtained compounds were identified by physicochemical properties and spectra data. Result: Twelve compounds were isolated from PE and Et OAc extracts of L. nervosa and their structures were identified as p-hydroxybenzoic acid( 1),protocatechuate( 2),3-( 4-hydroxyphenyl)-2-propenoic acid( 3),( E)-ferulic acid docosyl ester( 4),( Z)-ferulic acid docosyl ester( 5), stigmast-4-ene-3-one( 6), meranzin hydrate( 7), xanthotoxol( 8), auraptenol( 9),stigmasterol-3-O-β-D-glucoside( 10), vanillic acid( 11) and p-hydroxybenzaldehyde( 12). Conclusion:Compounds 1-10 were obtained from the genus Liparis for the first time,and compound 11 was obtained from L. nervosa for the first time.
