Synthesis of Related Substances of Canagliflozin

摘要 根据卡格列净的结构及合成工艺,制备了可能存在的4个工艺杂质:(1S)-1,5-脱氢-1-C-[3-[(5-苯基-2-噻吩基)-甲基]-4-甲基苯基]-D-葡萄糖醇(有关物质A)、(1R)-1,5-脱氢-1-C-[3-[[5-(4-氟苯基)-2-噻吩基]甲基]-4-甲基苯基]-D-葡萄糖醇(有关物质B)、(2R,3R,4R)-2-[(S)-1,2-二羟基乙基]-5-[3-[[5-(4-氟苯基)-2-噻吩基]甲基]-4-甲基苯基]四氢呋喃-3,4-二醇(有关物质C)及(2S,3R,4R,5R)-1-[3-[[5-(4-氟苯基)-2-噻吩基]甲基]-4-甲基苯基]己烷-1,2,3,4,5,6-六醇(有关物质D),并经MS、~1H NMR和^(13)C NMR等确证结构。 According to the structure and synthetic process of canagliflozin, four related substances: (IS)-1,5- dehydro-l-C-[3-E(5-phenyl-2-thienyl)methyl]-4-methylphenyl]-D-glucitol (A), (1R)-l,5-dehydro-l-C-[3-EE5-(4- fluorophenyl)-2-thienyl]methyl]-4-methylphenyl]-D-glucitol (B), (2R,3R,4R)-2-L(S)-l,2-dihydroxyethyl]-5-[3-[[5- (4-fluorophenyl)-2-thienyl]methyl]-4-methylphenyl]tetrahydrofuran-3,4-diol (C) and (2S,3R,4R,5R)-I-E3-[[5-(4- fluorophenyl)-2-thienyl3methyl]-4-methylphenyl]hexane-l,2,3,4,5,6-hexaol (D) were prepared, and their structures were confirmed by MS, 1H NMR and 13C NMR.
