摘要
目的:研究荷叶碱固体脂质纳米粒的制备处方及工艺。方法:以包封率为参考指标来评价乳化—超声法制备的荷叶碱固体脂质纳米粒(Nuc-SLN)的质量,单因素筛选的最佳处方,L_9(3~4)正交试验筛选出的最佳工艺组合。结果:单因素筛选的最佳处方:荷叶碱10mg,单硬脂酸甘油酯150mg,泊洛沙姆125mg;L_9(3~4)正交试验筛选出的最佳工艺组合为A_3B_2C_3D_1,即制备过程中搅拌速度1000 rpm/min,加热温度78℃,加热搅拌时间16 min,超声时间10 min。结论:采用最优处方及最佳工艺组合制备得到荷叶碱固体脂质纳米粒比较透明且有淡蓝色乳光,包封率和载药量较高,本研究方法可作为制备荷叶碱固体脂质纳米粒的制备方法。
Objective: To optimize the preparation formulation and technology of Nuciferin Solid Lipid Nanoparticles. Method: According to the reference index of entrapment rate to evaluate the quality of nuciferine solid lipid nanoparticles (Nuc-SLN) which were prepared by ultrasonic emulsification.Single factor (or univariate) inspect the prescription of Nuc-SLN. Use L(34) orthogonal test method to improve the preparation technology. Result: Through the single factor inspection of Nuc-SLN, the best prescription is: Nu- ciferinl0mg, Glyceryl Monostearatel50mg, Poloxamer125mg; Under orthogonal test , the best Nuc-SLN preparation technology is A3B2C3D3 , which is blend speed 1000 rpm/min, heating temperature 78℃ , heating time 16 min,uhrasonic time 10 rain. Conclusion: The nuciferine solid lipid nanopartieles with more transparent and light blue milk were prepared by combination of optimal prescrip- tion and the best technics, with high entrapment efficiency and drug loading, and this method can be used for preparing nuciferine sol- id lipid nanoparticles.
