柑橘素通对大鼠缺血再灌注心脏损伤作用后氧自由基和热休克蛋白70水平的影响 被引量:1

摘要 目的探讨柑橘素(Nari)对大鼠缺血再灌注心脏损伤作用后保护作用。方法将本研究中心56只雄性成年SD大鼠随机分为4组:对照组、Nari组、Nari+格列苯脲(Gli)组、Nari+5-羟基葵酸(5-HD)组;各组大鼠心脏给予30 min全心缺血、60 min再灌注;记录不同Nari浓度水平对再灌注心脏损伤作用,并评估其对氧自由基、热休克蛋白70水平的影响。结果心脏缺血/再灌注后60 min,2.5μmol/L以上Nari组大鼠心脏功能恢复优于对照组;Nari+Gli、Nari+5-HD大鼠心肌梗死面积、冠脉流出液LDH含量均高于10μmol/L Nari组大鼠;心脏缺血/再灌注后60 min,2.5μmol/L、5.0μmol/L、10.0μmol/L、20.0μmol/L Nari组心肌组织HSP70表达水平、SOD水平高于对照组,呈逐渐增加的趋势,心肌组织MDA水平低于对照组,呈逐渐降低的趋势,差异均有统计学意义(P<0.05)。结论柑橘素对心肌缺血再灌注心脏损伤具有保护作用。 Objective To investigate the protection of citrus( Nari) on the ischemia-reperfusion heart injury in rats. Methods 56 male SD rats in our study center were randomly divided into 4 groups: the control group,Nari group,Nari + glibenclamide( Gli)group,Nari + 5-hydroxycaproic acid( 5-HD) group; the hearts of rats in each group were given 30 min ischemia,60 min reperfusion. Effects of different concentrations of Nari on reperfusion injury in heart were recorded to evaluate their effects on oxygen free radical and heat shock protein 70 levels. Results Cardiac function was significantly better than that in the control group at 60 min after ischemia/reperfusion and 2. 5 μmol/L Nari rats. The infarct size and the LDH content of coronary flow of Nari-Gli and Nari-5-HD rats were higher than that of 10 μmol/L Nari rats. The expression level of HSP70 in myocardium and the level of SOD were higher than those of the control group at 60 min,2. 5 μmol/L,5. 0 μmol/L,10. 0 μmol/L and20. 0 μmol/L Nari,respectively,showing an increase trend,and the level of MDA in myocardium was lower than that in control group,showing a decrease trend,with the differences statistically significant( P〈0.. 05). Conclusion Citrusin has a protective effect on myocardial injury induced by myocardial ischemia and reperfusion.
