摘要
对纤维素、半纤维素分解菌MYB3和YB1菌株在不同温度和pH值条件下的存活性、酶活性及对秸秆分解特性进行初步研究。结果表明:MYB3菌株存活的最适条件为35℃,pH值8时;YB1菌株为30℃,pH值9。2种菌在发酵中生成的纤维素酶活性最高,分别达到19.28U/mL(MYB3-玉米秸秆-3d)和19.5U/mL(YB1-稻草-3d);半纤维素酶最高分别达到6.66 U/mL(MYB3-玉米秸秆-2d)和17.17 U/mL(YB1-稻草-4d);淀粉酶活性最高分别达到9.84U/mL(MYB3-玉米秸秆-3d)和10.65U/mL(YB1-稻草-5d)。MYB3菌株适合玉米秸秆发酵,纤维素、半纤维素分解率(5d)分别为39.03%和14.67%;YB1菌株适合稻草发酵,纤维素、半纤维素分解率(5d)分别为31.53%和37.44%。
The biological characteristics of the cellulolytic and hemicellulolytic microbes MYB3 strain and YB1 stain were studied in this research,including the decomposition characteristics for the straw and the survivability in different temperature and pH value.The results showed that the optimum survival conditions is 35℃,pH 8 for the MYB3 stain,and 30℃and pH 9 for the YB1 strain.The highest cellulase activity of the two kinds of bacteria was acquired under the fermentation condition,and the level was 19.28 U/mL(MYB3 corn stover 3 d)and 19.5 U/mL(YB1 rice straw 3 d).The highest hemicellulase activity reached 6.66(MYB3 corn stover 2 d),17.17 U/mL(YB1 rice straw 4 d).The highest amylase activity reached 9.84 U/mL(MYB3 corn stover 3 d),10.65 U/mL(YB1 rice straw 5 d).The MYB3 strain was suitable for corn stover fermentation,which the cellulose and hemicellulose decomposition rate(5 d)was39.03% and 14.67%.The YB1 strain was suitable for rice straw fermentation,which the cellulose and hemicellulose decomposition rate(5 d)were 31.53% and 37.44%.
