UPLC法测定α-倒捻子素对芒果炭疽病菌麦角甾醇含量的影响 被引量:1

Evaluation of the Effects of Alpha-mangostin against Ergosterol Content in Colletotrichum gloesporioides by UPLC

摘要 [目的]探明α-倒捻子素对芒果炭疽病菌麦角甾醇合成的影响。[方法]通过测定α-倒捻子素对芒果炭疽病菌菌丝生长毒力,建立超高效液相色谱法(UPLC)分析其对菌丝麦角甾醇含量的影响。[结果]采用UPLC法检测,麦角甾醇的保留时间为2.58 min,在2.5~100 mg/L时线性相关系数为0.9999,平均回收率为85.72%,相对标准偏差为4.24%。α-倒捻子素对芒果炭疽病菌菌丝生长的EC_(50)和EC_(80)值分别为73.01、403.96 mg/L;在生长抑制剂量下,α-倒捻子素显著降低病原菌中麦角甾醇含量(P<0.05),且对麦角甾醇合成抑制作用呈现剂量依赖性。[结论]UPLC法适用于麦角甾醇的快速检测,α-倒捻子素对芒果炭疽病菌生长和麦角甾醇合成具有明显抑制作用。 [Aims] The aims were to investigate the effects of α-mangostin on ergosterol of Colletotrichum gloesporioides. [Methods] The toxicity bioassay of antifungal agent was tested by mycelial growth rate method. The ergosterol extracted from C. gloesporioides was analyzed by UPLC method. [Results] The retention time for ergosterol was 2.58 min, the linear correlation coefficient was 0.9999 when the concentration ranged from 2.5 to 100 mg/L, the average recovery was 85.72% and the relative standard deviation was 4.24%. α-Mangostin strongly inhibit the mycelia growth with the EC_(50) and EC_(80) values of 73.01 and 403.96 mg/L, respectively. Exposed to a series of inhibitory concentrations of α-mangostin, the ergosterol contents of mycelium were reduced. In addition, α-mangostin inhibited ergosterol biosynthesis in a dose-dependent manner. [Conclusions] α-Mangostin evidently inhibited mycelial growth and the ergosterol biosynthesis of C. gloesporioides, the established UPLC method is suitable for fast determination of ergosterol.
