摘要
目的探讨子宫肌层组织内SULT2A1及SULT1E1的表达水平与子宫肌瘤发病的关系。方法选取2015年1月-2016年10月于沧州市沧县医院因患子宫肌瘤而行子宫全切除术或子宫次全切除术患者32例为子宫肌瘤组,并选取同年龄段的15例因子宫脱垂行子宫切除术患者作为对照组,采用化学发光分析法分别测定所有纳入患者血清及组织匀浆中雌二醇(E_2)、孕酮(P)含量,并以Western印迹方法检测SULT2A1蛋白的表达,最后采用Pearson相关分析分析组间子宫肌瘤发病与子宫肌层组织内SULT2A1及SULT1E1的相关性。结果 (1)肌瘤组织E_2、P含量显著高于肌瘤包膜外子宫肌层组及子宫脱垂组,肌瘤组血清E_2、P含量明显低于健康女性组(P<0.05);(2)SULT2A1、SULT1E1在肌瘤包膜外肌层组织、肌瘤组织、正常子宫肌层中表达存在显著差异(P<0.05),具体表现为膜外肌层组织<肌瘤组织<正常子宫肌层;(3)皮尔逊相关性分析显示,肌瘤组织SULT2A1、SULT1E1与相应组织组织匀浆中E_2、P水平呈显著负相关(P<0.05),而与血清中E_2、P水平无明显相关性。结论子宫肌瘤的发生、发展与子宫肌瘤组织局部SULT2A1、SULT1E1蛋白活性降低有关。
Objective To explore the relationships between expression levels of SULT2 A1 and SULT1 E_1 in myometrium and uterine fibroids. Methods From January 2015 to October 2016,32 patients with uterine fibroids( uterine fibroid group) underwent total hysterectomy or subtotal hysterectomy in the hospital,and 15 patients with uterine prolapse( uterine prolapse group) underwent hysterectomy. Chemiluminescence analysis was used to detect the levels of estradiol and progesterone in serum and tissue homogenate of patients in the two groups,Western blotting was used to detect the expression of SULT2 A1 protein,Pearson correlation analysis was used to analyze the correlations between onset of uterine fibroids and SULT2 A1 and SULT1 E_1 in myometrium. Results The levels of estradiol and progesterone in uterine fibroid tissue was statistically significantly higher than those in myometrium tissue out of fibroid membrane in uterine fibroid group and uterine prolapse group,the levels of estradiol and progesterone in uterine fibroid group were statistically significantly lower than those in healthy group( P0. 05). There were statistically significant differences in the expression levels of SULT2 A1 and SULT1 E_1 among myometrium tissue out of fibroid membrane,uterine fibroid tissue,and normal myometrium tissue( P0. 05),the expression levels in myometrium tissue out of fibroid membrane,uterine fibroid tissue,and normal myometrium tissue increased gradually. Pearson correlation analysis showed that the expression levels of SULT2 A1 and SULT1 E_1 in uterine fibroid tissue were significantly correlated with the levels of estradiol and progesterone in tissue homogenate( P0. 05),but they were not correlated with the levels of estradiol and progesterone in serum. Conclusion The occurrence and development of uterine fibroids is correlated with the decrease of SULT2 A1 and SULT1 E_1 protein activity in local uterine fibroid tissue.
