摘要
目的:探讨NOS/NO在白藜芦醇抗大鼠心肌缺血再灌注损伤中的作用。方法:健康雄性SD大鼠30只,随机分为3组(假手术组、缺血再灌注组、白藜芦醇组),采用结扎冠状动脉左前降支制备心肌缺血再灌注损伤模型,于结扎前15min及再灌注前1min经舌下静脉注射白藜芦醇10mg/kg,测定再灌注120min后血浆中NO含量和总NOS活性。结果:与假手术组和缺血再灌注组相比,白藜芦醇组NO含量明显升高(P<0.01),T-NOS活性明显增加(P<0.01);与缺血再灌注组比较,白藜芦醇组iNOS水平降低(P<0.05),而cNOS水平升高(P<0.01)。结论:白藜芦醇可能通过提高总NOS活性和增加NO含量产生对大鼠心肌缺血再灌注损伤的保护作用。
Objective:To explore the role of NOS/NO in the protective effect of resveratrol against myocardial ischemiareperfusion injury in rats.Methods:A total of thirty healthy male SD rats were randomly divided into 3 groups:sham operation group,ischemia/reperfusion(I/R)group,and post-disposal treatment of resveratrol(RSVL)group.The myocardial ischemia-reperfusion injury model was induced by ligating the left anterior descending branch of coronary artery.In addition,resveratrol was injected intravenously into the sublingual vein at a dosage of 10 mg/kg,respectively 15 min before ligation and 1 min before reperfusion.The content of nitric oxide(NO) and the activity of total nitric oxide synthase(T-NOS)in plasma were detected 120 min after reperfusion.Results:Results:Compared with sham operation group and I/R group,the content of NO and the activity of T-NOS were significantly increased in RSVL group(P<0.01).The level of iNOS was decreased in RSVL group after reperfusion(P<0.05),while the level of cNOS was increased(P<0.01).Conclusion:Resveratrol may have a protection against myocardial ischemia-reperfusion injury in rats by increasing the content of NO and the activity of T-NOS in plasma.
