血浆miRNAs在儿童急性淋巴细胞白血病(ALL)不同疾病状态中的表达特点

Expression of Plasma miRNAs in Children with Acute Lymphoblastic Leukemia in Different Disease States

目的分析3种血浆miRNA在儿童急性淋巴细胞白血病(ALL)不同疾病状态中的表达特点。方法选取急性淋巴细胞白血病患儿40例,其中初诊患儿12例,完全缓解患儿16例,复发患儿12例,同时收集我院经健康体检的儿童25例作为正常对照组。抽取所有儿童的空腹静脉血,离心后取血浆,采取直接扩增目的 microRNA,并反转录为c DNA,qRT-PCR方法检测血浆miR150、miR155、miR233的表达。结果 ALL组中血浆miR150、miR155、miR233的表达均与正常组有统计学差异;血浆miR150和miR233在初诊组和复发组的表达显著低于缓解组和正常组(P<0.05),血浆miR155在初诊组和复发组的表达显著高于缓解组和正常组(P<0.05)。3种miRNA在初诊组与复发组之间、缓解组与正常组之间比较均无统计学差异(P>0.05);初诊组中3种血浆miRNA在不同年龄和性别之间表达均无统计学差异(P>0.05);3种血浆miRNA的AUC(曲线下面积)和95%CI(95%可信区间)均有较好的满意度,且3种血浆miRNA联合检测的精确度更高。结论血浆miR150、miR155、miR233的表达在儿童急性淋巴细胞白血病(ALL)的诊断、治疗和复发监测中有较高的应用价值。

Objective To analyze the expression of 3 plasma miRNA in children with acute lymphoblastic leukemia in different disease states .Methods 40 children with acute lymphoblastic leukemia were collected ,including 12 cases of newly di-agnosed children,16 cases of complete remission,12 cases of relapse.At the same time,25 healthy children received physical ex-amination was the control group .The fasting venous blood was extracted from all the children .After centrifugation ,the plasma was taken directly to amplify the target microRNA ,and the reverse transcription into cDNA .QRT-PCR method was adopted to detect the expression of miR150,miR155 and miR233 in plasma.Results The expression of miR150,miR155 and miR233 in all group were significantly different from that of the control group .The expression of plasma miR150 and miR233 in the newly diagnosed and recurrent group were significantly lower than that in the remission group and the control group （P〈0.05）.The expression of plasma miR155 in the initial and recurrent group was significantly higher than that in the remission group and the normal group （ P〈0.05）.There was no significant difference in the 3 microRNA between the initial diagnosis group and the recurrence group ,the remission group and the control group （P〉0.05）;Three kinds of plasma miRNA in newly diagnosed group between different age and gender expression showed no significant difference （P〉0.05）;Three plasma AUC microRNA（area under the curve） and 95%CI（95% confidence interval ） had better satisfaction .And the accuracy of the combined detection of plasma miRNA in 3 kinds of plasma is higher .Conclusion The expression of plasma miR150,miR155 and miR233 has a good application value in the diagnosis ,treatment and recurrence monitoring of children with acute lymphoblastic leukemia .
