宫颈癌中自噬标记蛋白Beclin1、p62的表达及其临床意义 被引量:16

摘要 目的研究宫颈癌中自噬标记蛋白Beclin1、p62表达的水平及其对临床诊断及治疗的意义。方法选择子宫颈鳞状细胞癌组织标本140例,对纳入的组织标本使用Beclin1和p62蛋白标记。阳性结果采用半定量评分方法进行判定。Beclin1、p62的表达与患者临床特征相关性分析采用χ~2检验。结果 Beclin1主要表达于细胞膜及细胞质,与正常子宫颈上皮相比,Beclin1在宫颈癌组织中的表达明显降低(P<0.05)。但宫颈癌中Beclin1表达量与患者临床病理特征无相关性(P>0.05)。p62在子宫颈鳞状细胞癌组织细胞质中的表达明显升高,且与淋巴结的转移密切相关(P<0.05)。Beclin1和p62的阳性表达率之间未见明显的相关性(P>0.05)。结论 Beclin1、p62的表达量与子宫颈癌的发生发展相关,且p62的核浆转移与子宫颈癌有相关性。临床上,可根据Beclin1和p62的表达对疾病进展进行评估。 Objective To study the expression of autophagy marker protein Beclin 1,p62 and their significance for clini-cal diagnosis and treatment .Methods 140 cases of cervical squamous cell carcinoma specimens were selected ,tissue samples u-sing the included Beclin1 and p62 protein labeling.Positive results using semi-quantitative scoring method determination .Beclin1, p62 expression and clinical characteristics correlation analysis using χ2 test.Results Beclin1 mainly expressed in the cytoplasm and cell membrane ,as compared with normal cervical epithelium ,Beclin1 expression in cervical cancer was significantly lower ( P〈0.05).But Beclin1 expression and clinicopathological had no correlation (P〉0.05).Expression of p62 in cervical squamous cell carcinoma was significantly higher in the cytoplasm ,and was closely related with lymph node metastasis (P〈0.05).Beclin1 and positive expression of p62 had no significant correlation(P〉0.05).Conclusion The expression of Beclin1,p62 are related with development of cervical cancer ,and p62 nuclear transfer plasma is correlated with cervical cancer .Clinically ,the disease pro-gression can be assessed based on the expression of p 62 and Beclin1.
