摘要
目的观察当归补血汤与四物汤对血虚模型小鼠补血作用以及两个方剂的差异性。方法ICR小鼠40只,随机分成正常组、模型组、当归补血汤组和四物汤组,每组10只,除正常组外,另三组复制失血性血虚小鼠模型,当归补血汤组和四物汤组同时分别灌胃当归补血汤和四物汤,连续给药7天,观察小鼠体征特点,测定各组小鼠给药前及造模给药后血红细胞(RBC)、血红蛋白(HGB)、红细胞压积(HCT)、血小板计数(PLT)和白细胞(WBC)的值以及第7天出血凝血时间变化。结果给药7天,当归补血汤组小鼠食欲、行为和毛色等体征特点改善直至完全恢复,四物汤组小鼠体征特点改善不明显。与正常组比较,模型组小鼠各指标均降低[RBC:(6.65±0.87)×10^(12)/L比(9.90±0.95)×^(12)/L(P<0.01),HGB:(108.6±11.69)g/L比(134.4±13.55)g/L(P<0.01),HCT:(44.37±4.72)%比(52.01±4.98)%(P<0.01),PLT:(983±7.81)×109/L比(1218±10.27)×^(9)/L(P<0.01),WBC:(6.20±0.22)×^(9)/L比(7.80±0.05)×^(9)/L(P<0.01)];与模型组比较,当归补血汤组与四物汤组小鼠各指标均升高[当归补血汤组:RBC(9.31±0.89)×^(12)/L,HGB:(136.1±11.88)g/L,HCT(53.10±5.24)%,PLT(1005±10.43)×^(9)/L,WBC(8.26±0.31)×^(9)/L,P<0.05,P<0.01;四物汤组:RBC[(9.68±0.99)×^(12)/L,HGB(132.4±14.62)g/L,HCT(53.18±5.89)%,PLT[(1097±6.97)×^(9)/L,WBC(7.63±0.05)×^(9)/L,P<0.05,P<0.01]。结论当归补血汤和四物汤均有较好的补血作用,但两方对模型小鼠的血虚体征特点影响不同。
Objective To study the effect of Angelicae Sinensis Decoction and Siwu Decoction on blood deficiency model mice and the difference of two prescriptions. Methods Forty ICR mice were randomized into blank,model, Angelicae Sinensis Decoction, and Siwu Decoction groups, with 10 mice in each group. Hemorrhagic blood deficiency models were established in other 3 groups except for blank group. The treatment groups were fed with the corresponding drugs for consecutive 7 days. Physical signs were daily observed; the value of red blood cell( RBC), hemoglobin( HGB), hematocrit( HCT), platelet( PLT), white blood cell( WBC) were measured before and on day 1, 3 and 7 after treatment; the blood coagulation time was determined on day 7. Results On day 7 after treatment, the appetite, the behavior, and the appearance of mice in Angelicae Sinensis Decoction group was great-ly improved to the normal level, but such improvement was not noted in Siwu Decoction group. Compared with blank group, the RBC, HGB, HCT, PLT, and WBC levels of mice in model group were significantly lower respectively [RBC:( 6.65±0.87) ×^(12)/L vs( 9.90±0.95) ×^(12)/L; HGB:( 108.6±11.69) g/L vs( 134.4±13.55) g/L; HCT:( 44.37±4.72) % vs( 52.01±4.98) %; PLT:( 983±7.81) ×^(9) L vs( 1218±10.27) ×^(9)/L; WBC:( 6.20±0.22) ×^(9) L vs( 7.80±0.05)×^(9)/L; all P<0.01], but the reduction was reversed in both Angelicae Sinensis Decoction [RBC:( 9.31±0.89) ×^(12)/L;HGB:( 136.1±11.88) g/L; HCT:( 53.10±5.24) %; PLT:( 1005±10.43) ×^(9) L; WBC:( 8.26±0.31) ×^(9) L] and Siwu Decoction groups [RBC:( 9.68±0.99) ×^(12)/L; HGB:( 132.4±14.62) g/L; HCT:( 53.18±5.89) %; PLT:( 1097±6.97) ×^(9) L; WBC:( 7.63±0.05) ×^(9) L], with P <0.05 or P <0.01. Conclusion Angelicae Sinensis Decoction and Siwu Decoction have good blood enriching effect, but the two kinds of prescriptions have different effects on the characteristics of blood deficiency signs in model mice.
