高产漆酶白腐真菌的筛选 被引量:2

Screening of White Rot Fungi with High Laccase Yield

摘要 以6种白腐真菌作为供试菌株,5种食用菌作为对照菌株,采用愈创木酚及α-萘酚基质平板显色法,依据漆酶催化氧化还原反应产生显色圈的直径大小及变色程度对产漆酶菌株进行初筛;并采用ABTS酶活力测定法对菌株进行复筛。结果表明,11种真菌均具有分泌漆酶的能力,得到5株高产漆酶的白腐真菌菌株,由强至弱排列依次为朱红栓菌[Trametes cinnabarina(Jacq.∶Fr.)Fr]、木蹄层孔菌[Pyropolyporus fomentarius(L.Ex F.)Teng]、单色云芝[Coriolus unicolor(L.∶Fr.)Pat]、轮纹韧革菌[Stereum ostrea(Bl.et Nees)Fr.]和宽鳞棱孔菌[Favolus squamosus(Huds.ex Fr.)Ames],酶活力分别为23.47、20.26、19.52、18.39、17.93 U/m L。 With 6 kinds white rot fungi as test strains and 5 kinds edible fungi as control strain,the method of plate sub-strate chromogenic of Guaiacol and 琢- naphthol were adopted, the diameter of chromogenic and the degree of discoloration were compared 袁 the secrete laccase strains were screened preliminary. And strains were screened for the second time by the method of ABTS laccase activity determination. The results showed that all kinds of fungi had the ability to secrete laccase. 5 strains of high yield laccase white rot fungi were obtained,and the rank (from high to low) of these 6 kinds of fungi on the laccaseactivity were Trametes cinnabarina袁 Pyropolyporus fomentarius 袁 Coriolus unicolor ,Stereum ostrea and Favolus squamosus 袁 enzyme activities were 23.47 U/mL, 20.26 U/mL, 19.52 U/mL, 18.39 U/mL, 17.93 U/mL, respectively.
