摘要 目的:探讨核分裂周期蛋白80(nuclear division cycle 80,NDC80)在原发性肝细胞肝癌中的作用。方法:应用实时聚合酶链式反应(real-time polymerase chain reaction,real-time PCR)的方法分析肝细胞肝癌和对应的癌旁组织中NDC80的表达情况;将肝癌细胞株Hep3B转染慢病毒沉默NDC80基因的表达,通过real-time PCR和Western Blot的方法证实。克隆形成实验用来评估NDC80基因沉默对Hep3B细胞增殖的影响。结果:肝癌组织中NDC80的表达水平明显高于相应的癌旁组织。功能研究表明NDC80的沉默显著减少了Hep3B细胞的增殖和克隆形成。结论:NDC80的高表达可能在促进肝细胞肝癌的发展方面发挥作用。 Objective :To investigate the role of nuclear division cycle 80(NDC80) in human hepatocellular carcinogenesis. Methods :NDC80 gene expression was analyzed by real-time polymerase chain reaction(real-time PCR) in hepatoeellular carcinoma and adjacent tissues. The hepatocellular carcinoma cell line Hep3B was transfected with lentivirus to silence endogenous NDC80 gene expression, which was confirmed by real-time PCR and Western Blot. The effects of NDCS0 silencing on Hep3B cell proliferation were evaluated by the colony formation experiment. Results :NDC80 expression levels in hepatocellular carcinoma tissues were significantly higher than those in the adjacent tissues. Functional studies demonstrated that NDC80 silencing significantly reduced Hep3B cell proliferation and colony formation. Conclusion: Elevated expression of NDC80 may play a role in promoting the development of hepatocellular carcinoma.
