摘要 目的:探讨原发性肝癌组织中鞘氨醇激酶1(sphingosine kinase 1,SPHK1)的表达及其临床意义。方法:采用Real-time PCR和免疫组化检测252例原发性肝癌患者癌组织和癌旁组织中SPHK1的表达水平,分析其对肝癌患者生存和复发的影响。结果:SPHK1蛋白在肝癌组织中阳性表达率为71.8%,而在癌旁组织中为11.1%(P<0.01),高表达SPHK1蛋白的肝癌患者生存期较短(P=0.000)而复发率较高(P=0.025)。结论:SPHK1是肝癌的一个潜在分子靶点,有助于判断患者的复发和预后。 Objective:To investigate the expression of sphingosine kinase 1(SPHK1) in primary hepatocellular carcinoma tissues and its clinical significance. Methods: Real-time PCR and immunohistochemistry methods were used to detect the expression levels of SPHK1 in 252 cases of hepatocellular carcinoma tissues and paired normal tumor-adjacent tissues. The tumor recurrence and survival of the primary hepatocellular carcinoma patients were analyzed. Results:The positive expression rate of SPHK1 was 71.8% in primary hepatocellular carcinoma tissues,while 11.1% in paired adjacent tissues(P<0.01). The patients with high expression levels of SPHK1 protein had higher recurrence rate and poorer prognosis(P<0.01 or 0.05). Conclusion:SPHK1 may be a potential molecular target and helps to determine the patient's recurrence and prognosis.
