摘要
短毛五加总甙(AGVPS)是中药短毛五加(Acanthopanax gracilistylus var. pubescens)的有效成份。本文发现短毛五加总甙能抑制正常家兔由多种诱导因素所致的血小板聚集。同时,它还抑制花生四稀酸所致血小板聚集时TXA_2的产生,增加离体家兔胸主动脉PGI_2产生。在高脂喂养的动脉粥样硬化的家兔模型上,能提高血浆PGI_2水平。
We investigated the effect of the Chinese herb saponins of Acanthopanax gracilistylus var. pubescens (AGVPS) on platelet aggregation and PGI2 / TXA2 balance. AGVPS significantly inhibited in vivo rabbit platelet aggregation induced by ADP within 12 h after administering drugs, and also inhibited in vitro rabbit platelet aggregation induced by ADP, AA and collagen dose-dependently. AGVPS significantly inhibited in vitro TXB2 generation of the AA-stimulated rabbit platelets, and increased in vitro 6-keto-PGF1a generation of the rabbit thoracic aortae. In the atherosclerotic rabbits, AGVPS (60 mg ·kg -1·d-1 im for 6 wk) increased the plasma level of 6-keto-PGFla from 0.27±SD 0.13 ng/l to 1.08±0.78 ng/ml (p<0.05, n = 6) and decreased the plasma level of TXB2 from 0.52+ 0.27 ng/ml to 0.32±0.18 ng/ ml, but the differences were not significant. It is concluded that AGVPS inhibits platelet aggregation and regulates PGI2/TXA2 balance, which may result from the inhibition of TXA2 synthetase.
