响应面法优化红花蜂花粉多糖脱蛋白工艺 被引量:9

Optimization of Deproteinization Technology for Polysaccharides from Bee Pol en of Carthamus tinctorius by Response Surface Analysis

摘要 以蛋白质清除率和多糖保留率为评价指标,比较Sevage法、TCA法、酶法、Sevage-酶法和TCA-酶法的脱蛋白效率。确定PBPC脱蛋白最适方法为Sevage-酶法,即木瓜蛋白酶浓度2%,酶解时间2 h,pH 7.5,酶解温度50℃,Sevage法4次。该法蛋白清除率45.88%,多糖保留率86.49%。 Taking protein removal rate and polysaccharides retention rate as the evaluation indexes,the Sevage method,TCA method,enzyme method,Sevage-enzyme method and TCA-enzyme method were compared.Among the five methods,Sevage-enzyme method was the optimum deproteinization method.The deproteinization technologic condition was 2% enzyme concentration,pH of 7.5,enzymolysis temperature 50 ℃,and Sevage method repeat 4 times.Under this condition,deproteinized rate and polysaccharides retention rate were 45.88% and 86.49% respectively.
