摘要
目的改良冠脉结扎术制备大鼠慢性心衰模型。方法 60只SD大鼠随机分为假手术组10只和手术组50只,大鼠麻醉后,经口气管插管,连接呼吸机,开胸,结扎左侧冠状动脉前降支(假手术组只穿线不结扎),关胸缝合,6周后观察两组大鼠的心电图变化、存活率、心体比、血流动力学及心肌组织结构的变化。结果手术组50只大鼠6周后存活39只,存活率为78.00%;术后手术组心电图ST段明显抬高;6周后手术组较假手术组左室收缩压(LVSP)和最大上升速率(+dp/dt_(max))均下降,左室舒张末压(LVEDP)和最大下降速率(-dp/dt_(max))均上升(P<0.05);HE染色及心肌组织电镜可见手术组心肌结构发生明显变化。结论改良冠脉结扎术提高了术后大鼠的存活率,在一定程度上为成功制备慢性心衰模型提供了更大的可能。
Objective: To establish a rat model of chronic heart failure by modified coronary artery ligation. Methods: 60 SD rats were randomly divided into sham operation group(n = 10) and the operation group(n = 50). After anesthesia,the rats were intubated by mouth,connected to the ventilator,opened the chest and ligated the anterior descending branch of the left coronary artery (the sham group's line was not ligated),land ayer by layer closed chest. After 6 weeks, the changes of ECG, survival rate,ratio of heart to body, hemodynamics and myocar- dial tissue structure were observed in the operation group and sham operation group. Results: After 6 weeks, only 39 survived in 50 SD rats in the operation group;the survival rate was 78.00%. ECG of the operation group showed ST-segment elevation. After 6 weeks, compared with the sham group, both left ventricular systolic pressure (LVSP) and the maximum rise rate(+dp/dtmax) decreased in the Operation group ;both left ventricular end diastolic pressure(LVEDP) and the maximum decline rate(-dp/dtmax) increased(P〈 0.05). HE staining and myocardial tis- sue electron microscopy showed significant changes in myocardial tissue structure of the operation group. Conclu- sion: The improved coronary artery ligation improves the survival rate of rats after operation,and provides a greater possibility for the successful preparation of chronic heart failure models.
