五酯胶囊对万古霉素所致肾损伤大鼠的影响及其机制研究 被引量:3

Effect and Underlying Mechanism of Wuzhi Capsules in the Treatment of Vancomycin-induced Kidney Injury in Rats

摘要 目的:探讨五酯胶囊对万古霉素诱导所致肾损伤大鼠的影响及其可能机制。方法:24只雄性SD大鼠随机均分为空白对照组、模型对照组与五酯胶囊给药组,每组各8只。模型对照组与五酯胶囊给药组均连续每天1次尾静脉注射万古霉素200 mg·kg^(-1),空白对照组注射等剂量生理盐水,同时五酯胶囊给药组每天灌胃五酯胶囊混悬液0.25 g·kg^(-1),模型对照组与空白对照组则均灌胃等剂量蒸馏水。连续给药10 d后,检测大鼠血清中的胱抑素C(CysC)、肌酐(Scr)及尿素氮(BUN)水平、尿液中的24 h尿微量白蛋白(mAlb)、中性粒细胞明胶酶相关脂质运载蛋白(NGAL)及人肾损伤因子-1(KIM-1)水平及肾组织匀浆中的超氧化物歧化酶(SOD)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)水平,比较组间差异。结果:模型对照组与五酯胶囊给药组的肾指数、CysC、Scr、BUN、mAlb、NGAL、KIM-1及MDA水平均较空白对照组显著升高(P<0.05或P<0.01),且模型对照组升高幅度高于五酯胶囊给药组(P<0.05);模型对照组与五酯胶囊给药组的SOD、GSH-Px水平均显著低于空白对照组(P<0.05或P<0.01),且模型对照组下降幅度大于五酯胶囊给药组(P<0.05)。结论:五酯胶囊对万古霉素诱导所致肾损伤大鼠具有保护作用,可能与其抑制氧化应激作用有关。 Objective: To explore the protective effect of Wuzhi capsules on vancomycin-induced kidney injury in rats and investigate the action mechanism. Methods: Totally 24 male SD rats were randomly divided into the blank control group, the model control group and the model test group with 8 ones in each. The rats in the model control group and the model test group were intravenously injected vancomycin via tail veil, 200 mg·kg-1 per day for 10 consecutive days, while those in the blank control group were injected saline at the same volume. Meanwhile, the rats were orally administered Wuzhi solution (0.25 g·kg- l ) in the model test group and saline at the same volume in the blank control group and the model control group. The levels of 24-h microalbuminuria ( MALB), neutrophil getatinase associated lipocalin (NGAL) and kidney injury molecule 1 ( KIM-1 ) in urine were determined, and those of cystatin C (Cys C), serum creatinine (Scr) and blood urea nitrogen (BUN) in serum and those of superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) in renal tissues were also determined. Results: The levels of the renal index, Cys C, Scr, BUN, MALB, NGAL, KIM-1 and MDA in the model control group and the model test group were significantly higher than those in the blank control group, and those in the model control group were significantly higher than those in the model test group (P 〈 0.05 or P 〈0.01 ). The levels of SOD and GSH-Px in the model control group and the model test group were significantly lower than those in the blank control group, and those in the model control group were significantly lower than those in the model test group (P 〈 0.05). Conclusion: Wuzhi capsules can effectively relieve vancomycin-induced renal injury in rats probably by inhibiting the oxidative reaction.
