张家口坝上地区人群环氧合酶-2基因多态性与胃癌发病的相关性

Correlation of COX-2 genetic polymorphisms with incidence of gastric cancer in Bashang area of Zhangjiakou

摘要 目的分析张家口坝上地区人群环氧合酶-2(COX-2)基因多态性与胃癌发病的关系。方法选取2015年6月至2017年1月于张家口坝上地区居住10年以上的居民200例为研究对象,其中健康者100例作为对照组,胃癌患者100例作为胃癌组,采用聚合酶链反应-限制性片段长度多态性技术(PCR-RFLP)对两组进行基因分型,然后经非条件性Logistic回归分析COX-2基因单核苷酸多态性(SNP)rs3218625与胃癌发病风险的关系。结果对照组和胃癌组的年龄<65岁者比例分别为55.0%及58.0%,男性比例分别为70.0%及74.0%;连续或偶尔吸烟者比例为47.0%及54.0%,差异均无统计学意义(P>0.05);对照组和胃癌组COX-2 SNP rs3218625基因型为GG的比例分别为93.0%及89.0%;基因型为GA的比例分别为5.0%及10.0%;基因型为AA的比例分别为2.0%及1.0%,基因型为(AG+AA)的比例分别为7.0%及11.0%;经非条件性Logistic回归分析发现,COX-2基因SNP rs3218625与胃癌的发生密切相关,且基因型GA出现频率越高,胃癌发生风险越高(OR=2.463,P=0.021)。结论张家口坝上地区人群COX-2基因多态性可能会增加胃癌的发生风险,其中SNP rs3218625位点GA出现频率是胃癌发生的危险因素之一。 Objective To explore the correlation of cyclooxygenase-2 (COX-2) genetic polymorphisms with incidence of gastric cancer in Bashang area of Zhangjiakou. Methods A total of 200 residents who lived in Bashang ar-ea of Zhangjiakou for more than 10 years were selected from Jun. 2015 to Jan. 2017, including 100 healthy people (con-trol group) and 100 patients with gastric carcinoma (gastric carcinoma group). Polymerase chain reaction-restriction fragment length polymorphism technique (PCR-RFLP) was applied to analyze the genetic typing of the two groups, and the correlation of single nucleotide polymorphism (SNP) rs3218625 of COX-2 with the risk of gastric cancer was ana-lyzed by non-conditional logistic regression. Results The two groups showed no statistically significant difference in the proportion of subjects with age〈65 years (55.0%in the control group vs 58.0%in gastric carcinoma group), the pro-portion of males (70.0%vs 74.0%), and the proportion of subjects with continuous or occasional smoking history (47.0%vs 54.0%), P〉0.05. The proportion were 93.0% in the control group and 89.0% in gastric carcinoma group for GG of SNP rs3218625, 5.0%and 10.0%for GA, 2.0%and 1.0%for AA, and 7.0%and 11.0%for (AG+AA). Non-conditional logistic regression showed that COX-2 SNP rs3218625 was closely related with incidence of gastric cancer (OR=2.463, P=0.021). The higher the frequency of GA, the higher the risk of gastric cancer. Conclusion The COX-2 genetic poly-morphisms can increase the risk of gastric cancer in Bashang area of Zhangjiakou, and the frequency of GA in SNP rs3218625 locus is a risk factor for gastric cancer.
