川芎嗪对苯妥英钠血脑浓度以及脑内P-糖蛋白含量的影响 被引量:3

摘要 目的研究不同浓度川芎嗪对苯妥英钠血清、脑组织中浓度以及对脑内P-糖蛋白含量的影响。方法大鼠腹腔注射盐酸川芎嗪注射液协同灌胃苯妥英钠,采用高效液相色谱法检测大鼠血清及脑浆中苯妥英钠的浓度,采用酶联免疫法测定不同给药浓度的川芎嗪对脑内P-糖蛋白含量的影响。结果与苯妥英钠对照组相比,盐酸川芎嗪能显著增加血液及脑组织中的苯妥英钠浓度(P<0.05或P<0.01);降低脑组织中P-糖蛋白含量(P<0.01)。结论川芎嗪能够增加血液及脑组织中的苯妥英钠含量,降低脑组织P-糖蛋白的浓度含量,具有促进苯妥英钠入血和通过血脑屏障的作用。 Objective To determine the effect of different concentrations of tetramethypyrazine (TMP) on the concentration of phenytoin in serum and brain tissue and the effect on the content of P-glycoprotein in brain. Methods Clinically equivalent concentration of TMP, was intraperitoneally injected to rats, and the phenytoin was fed. The concentration of phenytoin sodium in the serum and the brain tissue of different groups was determined by HPLC. The concentrations of P-glycoprotein in the brain tissue of TMP at different drug concentrations was measured by enzyme linked immunosorbent assay (ELISA). Results @ Compared with the control group, the ligustrazine hydrochloride group markedly increased the concentration of phenytoin sodium in the blood and the brain tissue (P 〈 0.05 or P 〈 0.01). @ Compared with the control group, the ligustrazine hydrochloride group markedly reduced the content of P-glycoprotein in the brain tissue (P 〈 0.01). Conclusion Ligustrazine hydrochloride can increase the content of phenytoin sodium in the blood and the brain tissue, reduce the concentration of P-glycoprotein in the brain tissue, and promote the permeation of phenytoin into the blood and cross the blood brain barrier.
