摘要
目的:探讨H19多态性与肿瘤易感性之间的关系。方法:检索Pub Med,Embase与Web of Science数据库得到H19基因多态性与肿瘤易感性的病例对照研究,采用比值比(OR)和95%可信区间(95%Cl)评价关联强度。然后进行亚组分析、敏感性分析和文献发表偏倚检验。结果:最终纳入5篇病例对照研究,包括6 207例肿瘤患者和7 072例健康对照。分析后发现H19 rs217727多态性与肿瘤易感性无关(AA vs GG:OR=1.11,95%CI 0.83~1.48,P_h=0.01,I^2=69.5%;GA vs GG:OR=0.88,95%CI 0.73~1.06,P_h=0.02,I^2=65.0%;GA/AA vs GG:OR=0.94,95%CI 0.78~1.12,P_h=0.02,I^2=66.3%;AA vs GG/GA:OR=1.19,95%CI 0.93~1.53,P_h=0.02,I^2=64.9%)。同样地,按照亚组分析提示在亚洲人中存在相同结果。然而,H19rs2839698多态性与肿瘤易感性相关(TT vs CC:OR=1.30,95%CI 1.01~1.68,P_h=0.17,I^2=40.6%;TT vs CC/CT:OR=1.30,95%CI 1.07~1.59,P_h=0.31,I^2=15.7%)。结论:H19 rs217727多态性与肿瘤易感性无关,H19 rs2839698多态性与肿瘤易感性相关。
Objective:To evaluate the association between the H19 polymorphisms and the risk of cancer. Meth- ods:PubMed,Embase and Web of Science was searched to get the case - control study on the association between H19 polymorphisms and cancer susceptibility. Odds ratio (OR) and 95% confidence interval (CI) were calculated to evaluate the strength of the associations. Then subgroup analysis, sensitivity analysis, and publication bias were performed. Results : Five studies were included, involving a total of 6 207 cancer cases and 7 072 healthy controls. No sta- tistical relationship was observed between H19 rs217727 polymorphism and cancer risk (AA vs GG:OR = 1.11,95% CI 0.83 - 1.48, Ph = 0.01,12 = 69.5% , GA vs GG: OR = 0.88,95% CI 0.73 - 1.06, Ph = 0.02, Iz = 65.0% , GA/ AA vs GG: OR = 0.94,95% CI 0.78 - 1.12,Ph = 0.02,12 = 66.3 %, AA vs GG/GA : OR = 1. 19,95 % CI 0.93 1.53,Ph = 0.02,12 = 64.9% ). Similarly, subgroup analysis in Asian showed no significant association between rs217727 and cancer susceptibility. However, the H19 rs2839698 polymorphism was associated with risk of cancer ( TT vs CC : OR = 1.30,95% CI 1.01 - 1.68 ,Ph =0. 17 ,I^2 =40.6% , Tr vs CC/CT : OR = 1.30,95% CI 1.07 1.59,Ph = 0.31,12 = 15.7% ). Conclusion: The H19 rs217727 polymorphism might not be associated with overall cancer risk. However,The H19 rs2839698 polymorphism is associated with risk of cancer.
