摘要
为研究谷胱甘肽硫转移酶A3(GSTA3)对胫骨软骨发育不良(TD)肉鸡血清中凋亡因子Bax的影响,试验将90羽1日龄AA肉仔鸡预饲7 d后,随机分为6组(A,B,C,D,E和F组),分别在8,9日龄对D,E,F组饲喂添加100 mg/kg福美双的日粮,将饲喂基础日粮组(A,B,C组)和添加福美双日粮组(D,E,F组)的肉鸡各分对照组(A,D组)、重组GSTA3蛋白低剂量(20μg/kg)注射组(B,E组)和高剂量(50μg/kg)注射组(C,F组),在试验第8,10,12天腿部肌肉注射重组GSTA3蛋白,对照组注射稀释用PBS。在饲喂福美双后第1,2,4天分别采集各组肉鸡血液、分离血清,利用ELISA方法测定肉鸡血清中Bax的含量。结果显示,低剂量的重组GSTA3蛋白显著降低第2天基础日粮(B)组肉鸡血清中Bax的浓度,同时显著降低第4天福美双处理(E)组肉鸡血清中Bax的浓度;高剂量的重组GSTA3蛋白显著降低第4天基础日粮(C)组和福美双处理(F)组肉鸡血清中Bax的浓度。低剂量的重组鸡GSTA3蛋白对降低TD肉鸡血清中Bax含量的效果较好,这一研究将为肉鸡TD的防治提供新的方向和研究基础。
To investigate the effect of glutathione S-transferase A3(GSTA3)on the expression of apoptotic factor Bax in serum of tibial dyschondroplasia(TD),the study used 90 one-day-old AA broilers were divided into 6(A,B,C,D,E and F)groups at random after basal-diet feeding for 7 days.D,E and F of above 6 groups were fed with diets containing 100 mg/kg of thiram at 8 and 9 days old.Subsequently,feeding basal-diet(A,B and C)and adding thiram diets(D,E and F)were divided into control group(A and D),low dose(20 μg/kg)injection group(B and E)and high dose(50 μg/kg)injection group(C and F).The recombinant broiler protein GSTA3 was injected of leg intramuscularly at 8,10,12 days old,and the control group was injected with PBS for dilution.The blood collected after day 1,2 and 4 in TD broilers induced by thiram.The serum was isolated and the content of Bax was determined by ELISA.The results showed that the level of Bax in serum of broiler significantly decreased in basal-diet group(B)at 2 days and the level of Bax in serum of broiler significantly decreased in treated group(E)at 4 days because of the injection of low doses of GSTA3 recombinant protein(P0.05).The level of Bax in serum of broiler significantly decreased in basal-diet group(C)and treated group(F)at 4 days because of the injection of high doses of GSTA3 recombinant protein(P0.05).Low-dose recombinant protein GSTA3 has a better effect on reducing Bax content in TD broiler serum,and this study will provide a new direction and research basis for the prevention and treatment of TD.
