脱色菌群的富集及其对活性艳红X-3B的脱色效果 被引量:1

摘要 通过定向驯化法,从实验室活性污泥处理系统中,富集驯化出对活性艳红X-3B具有良好脱色性能的菌群H1~H5,针对其染料脱色性能进行了一系列研究。结果表明,脱色菌群有一定的耐盐能力,在盐度为15%,染料浓度100 mg·L^(-1)时,24 h后脱色菌群H5的脱色率为86%;脱色菌群H1~H5在低染料浓度下能够快速脱色(<600 mg·L^(-1)),在800~1 500 mg·L^(-1)的浓度范围内,经过48 h的培养后,脱色率都能达到95%以上;同时染料浓度为500 mg·L^(-1)时,脱色菌群H5脱色的适宜p H5~11,温度25~37℃,培养时间24 h,脱色率达到90%以上;脱色菌群H5对直接大红4BS和分散艳蓝KN-R有着良好的脱色性能,培养24 h,脱色率达到90%以上。利用扫描电镜观察,发现脱色菌群H5以杆菌为主。基于Illumina Miseq测序平台研究了脱色菌群H5的微生物菌群,发现其中优势菌属为Alcaligenes sp.和Pseudomonas sp.,所占比例分别为55.78%和11.14%。 The decolorizing bacterial consortium H1-H5 capable of decoloring reactive brilliant red X-3B ef- fectively were screened out from an activated sludge treatment in a laboratory by the directional domestication and their decolorization performances were studied. Results showed that the decolorizing bacterial consortium showed certain salt tolerance in decolorization. The bacterial consortium H5 could remain a decolorization rate of 86% under 15% salinity and 100 mg L-1 reactive brilliant red X-3B after 24 h. The decolorizing bacterial consor- tium H1-H5 could quickly decolor at a low concentration of dye (〈600 mg-L-1). In a range of 800-1500 mg.L-1, the decolorization rate could reach more than 95% after 48 h. At the same time, optimal conditions for the decol- orization of reactive brilliant red X - 3B were pH 5-11, 25-37℃ and culture for 24 h, and the decolorization rate was higher than 90% when the initial concentration of reactive brilliant red X-3B was 500 mg.L1. H5 had a good decolorization performance for direct scarlet 4BS and disperse brilliant blue KN-R, and the decolorization rate was higher than 90% after 24 h. Scanning electron microscope (SEM) revealed that the dominant bacteria were bacilli in the decolorizing bacterial consortium H5. Microbial community of the decolorizing bacterial consortium was investigated via Illumina MiSeq high throughput sequencing. The result indicated that Alcaligenes sp. (55.78%) and Pseudomonas sp. (11.14%) were two dominating bacteria.
