Yes相关蛋白在慢性鼻窦炎伴鼻息肉中的表达及与细胞增殖的相关性分析

Correlation analysis between expression levels of Yes-associated protein and cell proliferation in chronic rhinosinusitis with nasal polyps

目的检测Yes相关蛋白(YAP)在慢性鼻窦炎伴鼻息肉(CRSw NP)中的表达情况,初步探讨YAP在CRSw NP发病机制中的作用。方法收集对照组(13例)的下鼻甲黏膜和CRSw NP组(27例)的鼻息肉,采用实时荧光定量PCR的方法检测组织中YAP、TEAD1、TSLP、IL-33和Ki-67mRNA的表达情况,并分析相关性。结果鼻息肉组织中YAP、TEAD1、TSLP、IL-33和Ki-67 mRNA的相对表达量均高于对照组,组间差异有统计学意义(P均<0.05)。YAP mRNA相对表达水平分别与TEAD1、TSLP、IL-33、Ki-67 mRNA相对表达水平呈正相关(r_s=0.568;r_s=0.693;r_s=0.745;r_s=0.561;P均<0.01)。结论 YAP可能参与了CRSwNP的发病机制,并且可能与TSLP、IL-33等上皮因子的表达增加从而介导鼻息肉细胞增殖有关。

Objective To measure the expression levels of Yes-associated protein （YAP） in chronicsinusitis with nasal polyps （ CRSwNP） and preliminarily investigate the effect of YAP upon the pathogenesis of CRSwNP. Methods The inferior turbinates mucosa was collected in the control group （ n = 13） and nasal polyp tissue was obtained in the CRSwNP group （n = 27）. Fluorescent quantitative RT-PCRwas performed toquantitatively measure the expression levels of YAP, TEAD1,TSLP,IL-33 and Ki-67 mRNA, and analysis was conducted. Results The relative expression levels of YAP, TEAD1,TSLP, IL-33 and Ki-67mRNA in the CRSwNP tissues were significantly higher compared with those in the control group （all P 〈 0. 05 ）. The relative expression level of YAP mRNA was positively correlated with that of TEAD1 （ r = 0. 558, P=0.002）,TSLP （rs =0.693, P 〈0.001 ） ,IL-33 （ r s = 0.7 4 5 , P 〈 0 .0 0 1 ） and K i -7 mRNA （ r s = 0 .5 6 1,P 〈0. 001）. Conclusions YAP is probably involved with the pathogenesis of CRSwNP. It can mediate thecell proliferation of CRSwNP probably through up-regulating the expression levels of TSLP, IL-33 and other epi-thelial factors.
