五十八份菊芋种质资源遗传多样性SRAP分析 被引量:10

Genetic Diversity of Fifty-eight Jerusalem Artichoke Germplasm Resources Revealed by Sequence Related Amplified Polymorphism(SRAP)

摘要 以58份菊芋种质资源为试材,利用SRAP分子标记技术对其进行遗传多样性研究,为选育耐盐新品种提供参考依据。结果表明:从110对引物中筛选出16对多态性好的引物,共扩增出123条条带,其中多态性条带90条,多态性比率为72.7%。利用NTSYS软件统计分析出菊芋资源间遗传距离为0.01~0.52。应用UPGMA聚类可将58份菊芋资源划分为三大类群,第一大类群分为4个亚类群。表明利用SRAP技术更能充分揭示菊芋资源间的遗传差异,可作为种质鉴定依据。 Fifty-eight accessions of Jerusalem artichoke were used as materials,SRAP marker was used to analyze genetic diversities,in order to supply references for breeding salt-tolerant new variety. The results showed that 16 primers selected from 110 SRAP-primer were used for amplification and 123 DNA bands were obtained,aincluding 90 polymorphic bands. The percent of polymorphic was 72. 7%. The genetic distance indexes of 58 cultivars based on the SRAP data varied from 0. 01 to 0. 52. Cluster analysis with UPGMA method showed that the 58 resources tested could be divided into three groups.The first group could be divided into four sub groups. The results showed that The SRAP could fully reveal genetic differences for Jerusalem artichoke resources,and could be used in Jerusalem artichoke germplasms identification.
