摘要
目的:研究水飞蓟素胶囊促进大鼠胫骨骨折愈合的效果。方法:选取健康雌性SD大鼠72只,采用开放式截骨骨折模型,造模成功后随机分3组,对照组(CK组)每日灌服蒸馏水,观察组(SIL组)给予水飞蓟素100 mg·kg^(-1)·d-1灌胃,阳性对照组(JTG组)给予金天格0.48 g·kg^(-1)·d-1灌胃,在术后2,4,6周各组分别处死8只大鼠,行血清生化检测碱性磷酸酶(ALP),行离体胫骨骨矿物质密度(BMD)测定。组织行HE染色观察骨折愈合的病理变化,同时拍X线行影像学评估。结果:(1)血清ALP水平:在术后2周,表现出JTG组高于SIL组,并高于CK组(JTGvs.SIL,P<0.05;SIL vs.CK,P<0.05;JTGvs.CK,P<0.01)。但在第4、第6周,3组ALP含量无明显统计学差异(P>0.05)。(2)BMD值在3个时段均显示JTG组高于CK组(P<0.05),SIL组高于CK组(P<0.05),但JTG组与SIL组无统计学差异。(3)影像学评分:术后2周和4周,JTG组、SIL组高于CK组(P<0.05),但两组间无差异,而术后6周各组之间没有显著的统计学差异。(4)HE染色:在第2,4周,骨折愈合评分JTG组高于SIL组,并高于CK组(JTGvs.SIL,P<0.05;SIL vs.CK,P<0.05;JTG vs.CK,P<0.01),而术后6周3组间无明显统计学差异。SIL组早期即出现肉芽组织纤维化过程,并在相同的时间点其软骨细胞数量多于对照组。结论:水飞蓟素在早期具有明显的促进大鼠胫骨骨折愈合作用。
ORIECTIVE To study the healing effect of legalon capsules on fracture in rats. METHODS Seventy two female SD rats were enrolled in this study. The open osteotomy model was chosen. The rats were then randomly divided to three groups. Rats were fed with distilled water in control group (CK), with 100 mg-kg-1 .d-1 legalon in observation group (SIL) and with 0. 48 g.kg-a .d- a Jin Tiange in positive control group (JTG) after surgery. Eight rats of every group were killed at the 2nd, 4th ,6th weeks, serum alkaline phosphatase (ALP) and the isolated rat tibia bone mineral density (BMD) were meas- ured, the specimens were observed in histological examinations to observe the histological fracture healing, the bone callus and average healing scores were evaluated by radiographs. RESULTS The serum ALP showed that the JTG group was superior to SIL group and CK group (P〈0. 05) in 2nd week (JTG vs. SIL, P〈0. 05; SIL vs. CK, P〈0. 05; JTG vs. CK, P〈0. 01). But in 4th and 6th weeks, three groups showed no significant difference (P〈0. 05). The BMD values of JTG group and SIL group at each time point were higher than those of the CK group after operation (P〈0. 05). But there was no significant differ- ence between the JTG and SIL group. Bone callus and average healing scores: in the 2nd and 4th weeks after operation, JTG group and SIL group^CK group (JTG vs. SIL, P〈0. 05; JTG vs. CK, P〈0. 05; SIL vs. CK, P〈0. 05), and there was no significant differences among each groups in 6 weeks. HE staining showed that, in the 2nd and 4th weeks, JTG and SIL group were significantly faster than that of CK group (JTG vs. CK, P〈0. 01 ; SIL vs. CK, P〈0. 05 and JTG vs. SIL, P〈0. 05) ; but there was no significant differences among three groups in 6th week after operation. There was granulation tissue fibrosis on early stage in SIL group, and more cartilage ceils in SIL group than CK group at the same point of time. CONCLUSION The legalon capsules have significant effects in promoting rat tibial fracture healing in early time.
