摘要
目的探讨RAS相关区域家族6基因(RASSF6)和原癌基因K-RAS在胃癌组织中的表达及其临床意义。方法采用实时定量荧光PCR(qRT-PCR)检测20例新鲜胃癌组织和癌旁组织中RASSF6和KRASmRNA表达水平,应用免疫组化法检测100例胃癌组织和癌旁胃组织中RASSF6和K-RAS蛋白的表达水平。结果 RASSF6mRNA及其蛋白在胃癌组织中的表达低于癌旁组织,K-RASmRNA及其蛋白在胃癌组织中的表达高于癌旁胃组织(P均<0.05);分化度低、浸润度深、有淋巴结转移及TNM分期高的胃癌组织存在RASSF6低表达和K-RAS高表达(P均<0.05),胃癌组织中RASSF6与K-RAS的表达呈负相关(r=-0.269,P<0.01)。结论胃癌组织中存在RASSF6低表达和K-RAS的高表达;RASSF6在胃癌发生、发展中起负性调控作用。
Objective To investigate the expression and significance of RAS related region family 6 gene(RASSF6) and proto oncogene K-RAS in gastric cancer. Methods By using real-time quantitative PCR(qRT-PCR),mRNA level of RASSF6 and K-RAS in 20 cases of fresh gastric cancer tissues and adjacent normal tissues were analyzed. The expression level of RASSF6 and K-RAS protein were detected by immunohistochemical method in 100 cases of gastric cancer tissues and adjacent normal tissues. Results The expression of RASSF6 mRNA and protein in gastric cancer tissues were significantly lower than that in adjacent normal tissues. The expression of K-RAS mRNA and protein in gastric cancer tissues were higher significantly than that of adjacent normal tissues(P〈0.05). The low degree of differentiation,high invasion,and lymph node metastasis gastric cancer tissue had high expression of K-RAS and low expression of RASSF6(P〈0.05).The expression of RASSF6 was negatively correlated with K-RAS in gastric cancer tissues(r=-0.269,P〈0.01).Conclusion There were low expression of RASSF6 and high expression of K-RAS in gastric cancer tissues.RASSF6 played a negative regulatory role in the carcinogenesis and development of gastric cancer.
