摘要
目的:观察五味子乙素对胃癌SNU-1细胞增殖和转移的影响,探讨其诱导胶质瘤SNU-1细胞凋亡机制。方法:不同浓度五味子乙素组和顺铂组加入到SNU-1细胞中,作用24h后,MTT法测定药物组对胃癌SNU-1细胞增殖的抑制作用,流式细胞术检测不同浓度的五味子乙素诱导胃癌SNU-1细胞凋亡率,Transwell法检测五味子乙素对SNU-1细胞侵袭的抑制作用;Western blot法检测胃癌SNU-1细胞中Caspase-3、Bcl-2、Bax、E-cadherin、N-cadherin和MMP-9的表达,结果:与空白对照组比较,10、50和100μg/ml的五味子乙素和顺铂对胃癌SNU-1细胞生长的抑制作用明显,10、50和100μg/ml的五味子乙素作用胃癌SNU-1细胞后,细胞转移能力显著下降,10、50和100μg/ml的五味子乙素作用胃癌SNU-1细胞48h后均能诱导胃癌SNU-1细胞凋亡;10、50和100μg/ml的五味子乙素上调Caspase-3、Bax和E-cadherin的表达,下调Bcl-2和N-cadherin和MMP-9的表达。结论:五味子乙素可能是通过Caspase-3级联凋亡途径和线粒体途径诱导了胃癌SNU-1细胞凋亡,也可能通过破坏细胞间和基底膜粘附性和增加肿瘤细胞间的稳定性,来抑制胃癌SNU-1细胞的转移袭能力。
Objective:To investigate the related mechanism of Schisandrin B(Sch B) on proliferation and metastasis of Rhodiola glucoside for glioma cell.Methods:The inhibitory rate of Sch B on SNU-1 cells were determined using MTT method.The effect of Sch B on inhibiting metastasis of SNU-1 cells was determined by transwell assay.Finally the effect of expression of related proteins of SNU-1 cells was discussed.The SNU-1 cells apoptosis rate was detected by flow cytometry.Results:The growth inhibitory rates of SNU-1 cells were increased with the increasing of dose of salidroside(10,50,100μg/ml),and there were significant differenced.Apoptosis flow cytometry test results showed that SNU-1 cells apoptosis rate was increased with the increasing of dose of Sch B(10,50,100μg/ml).The expression of Sch B on Caspase-3,Bax,and E-cadherin of SNU-1 cells were increased,Bcl-2,N-cadherin,and MMP-9 of SNU-1 cells were decreased.Conclusion:Sch B may be reduced the inecreasd expression of Caspase-3,Bax and E-cadherin,and the decreased expression of N-cadherin and MMP-9.Eventually playes an important role on apoptosis of SNU-1 cells.
