摘要
目的建立苗药铁筷子的高效液相色谱(HPLC)特征指纹图谱,结合化学计量学方法对多个批次药材质量进行评价。方法采用Amethyst C_(18)-H色谱柱(4.6 mm×250 mm,5μm);0.1%甲酸水(A)-流动相为乙腈(B),梯度洗脱;流速0.8 m L·min^(-1),检测波长为254 nm,柱温28℃。通过对比保留时间和紫外吸收特征,对建立的指纹图谱的主要特征峰进行确认,采用相似度软件,结合系统聚类分析(HCA)和主成分分析(PCA)方法 ,对10个批次药材进行质量评价。结果建立了铁筷子HPLC指纹图谱,用对照品标定3个峰,相似度在0.978 0~0.991 9之间,10批药材亲缘关系较近。结论该方法专属性强,重现性良好,可为铁筷子的鉴别及质量评价提供依据。
OBJECTIVE To establish the high performance liquid chromatography (HPLC) fingerprint of the caulis of Chimo- nanthus nitens and evaluate the product quality by chemometrics analysis method. METHODS The method was developed on an Am- ethyst C ls-H column (4. 6 mm x 250 mm,5 μm)by gradient elution with acetonitrile-water (containing 0. 1% formic acid) as mobile phase at a flow rate of 0. 8 mL min-1. The column temperature was maintained at 28 %, and the detection wavelength was set at 254 nm. The main characteristic peaks was identified by comparing the retention time and UV absorption characteristics. Then 10 batches of the caulis of Chimonanthus nitens were evaluated by similarity assay, HCA, and PCA. RESULTS The HPLC fingerprint of the caulis of Chimonanthus nitens was established and three main peaks were identified. The similarity of 10 batches of the caulis of Chimo- nanthus nitens was about 0. 978 0 to 0. 991 9. CONCLUSION The established method can be used for the quality control of the caulis of Chimonanthus nitens.
