一种多功能营养复合物对四氧嘧啶诱导的实验性高血糖小鼠血糖及胰腺组织病理形态的影响 被引量:2

Effect of a multifunctional nutrient complex on blood glucose and histopathology of pancreas in alloxan-induced experimental hyperglycemia mice

摘要 目的观察一种多功能营养复合物对四氧嘧啶诱导的糖尿病小鼠血糖及胰腺组织病理形态的影响。方法将造模成功的昆明小鼠分为模型(MOD)组,高(HC)、中(MC)、低剂量(LC)复合物组和二甲双胍(MEL)组,同时选取正常昆明小鼠分为正常对照(Con)组和高剂量复合物对照(Hcon)组。灌胃45d后检测各组体重、FPG和血糖曲线下面积(AUCg)的变化,并测定血清胰岛素水平,观察胰腺组织病理形态。结果与Con组比较,Hcon组FPG[(5.88±0.52)vs(5.94±0.21)mmol/L]、AUCg[(14.23±1.85)vs(15.35±0.26)mmol/L]和血清胰岛素水平[(15.62±0.54)vs(15.01±0.99)mU/L]差异均无统计学意义(P>0.05)。与MOD组比较,HC、MC、LC组和MEL组FPG[(20.16±1.66)vs(11.78±1.25)vs(13.00±1.89)vs(13.79±1.37)vs(12.24±1.36)mmol/L,P<0.05)]和AUCg[(44.01±1.52)vs(32.49±2.73)vs(35.62±2.09)vs(37.39±1.88)vs(33.06±1.71),P<0.05)]降低,血清胰岛素含量增加[(8.99±0.68)vs(13.37±0.88)vs(12.86±0.63)vs(11.92±0.48)vs(14.29±0.78)mU/L,P<0.05)],高剂量复合物胰岛素敏感指数提高[(-5.50±0.21)vs(-4.91±0.13),P<0.05)]。胰腺组织切片染色结果显示,高、中、低剂量复合物和二甲双胍能够修复受损的胰岛组织,并且胰岛素表达量积分光密度值与MOD组比较升高[(77460±4849)vs(200400±11790)vs(127300±11750)vs(109900±19400)vs(194300±19400),P<0.05)]。结论该复合物能降低糖尿病小鼠FPG和AUCg,增加血清胰岛素含量,提高IS,可能主要是通过促进受损的胰岛β细胞修复实现的。 Objective To investigate the effect of a multifunctional nutrient complex on fasting blood glucose(FPG),area under the blood glucose curve(blood glucose AUCg),serum insulin level and histopathology of pancreatic in alloxan-induced experimental hyperglycemia mice. Methods The diabetic model mice were divided into five groups(MOD,HC,MC,LC,and MEL group),and ad ministrated orally with saline,high,medium and low doses of multifunctional nutrient complex and metfor min respectively.Healthy mice were also enrolled in this study and divided into two groups(Con and Hcon group).The body weight,FPG and blood glucose(AUCg)were measured at 45 d.The level of serum insulin was measured.Histopathology of pancreatic tissue were also evaluated. Results There were no significant difference in FPG [(5.88±0.52)vs(5.94±0.21)mmol/L],AUCg [(14.23±1.85)vs(15.35±0.26)mmol/L]and serum insulin level[(15.62±0.54)vs(15.01±0.99)mU/L]between Hcon group and Con group(P〉0.05).FPG [(20.16±1.66)vs(11.78±1.25)vs(13.00±1.89)vs(13.79±1.37)vs(12.24±1.36)mmol/L,P〈0.05]and AUCg[(44.01±1.52)vs(32.49±2.73)vs(35.62±2.09)vs(37.39±1.88)vs(33.06±1.71),P 〈0.05]decreased,while serum insulin level[(8.99±0.68)vs(13.37±0.88)vs(12.86±0.63)vs(11.92±0.48)vs(14.29±0.78)mU/L,P〈0.05]increased in HC,MC,LC and MEL group than in MOD group.Insulin sensitivity index increased in HC group than in MOD group[(-5.50±0.21)vs(-4.91±0.13),P 〈0.05].The hematoxylin-eosin staining results showed that the high,medium,low doses of complex and metfor min restored the damaged islet tissue,and significantly increased the integral optical density when compared with the diabetic model group[(77460±4849)vs(200400±11790)vs(127300±11750)vs(109900±19400)vs(194300±19400),P〈0.05].ConclusionThe multifunctional nutrient complex could reduce FPG and blood glucose AUCg,increase serum insulin,and improve insulin sensitivity in diabetic mice.The mechanism may be through promoting the repairment of damaged isletβcells.
