脂肪源性间充质干细胞移植治疗糖尿病大鼠勃起功能障碍的研究

摘要 目的探讨脂肪源性间充质干细胞(AD-MSCs)移植到糖尿病(DM)勃起功能障碍(ED)大鼠后,改善相关勃起功能的可能机制。方法制备、分离、培养AD-MSCs,研究其体内、外特性。新生雄性大鼠腹腔注射5-乙炔基-2-脱氧尿苷以跟踪内源性MSCs,8周后随机选择8只大鼠作为正常对照组(N组),其余大鼠以剂量60 mg/kg链脲佐菌素(STZ)行腹腔注射诱导DM,DM大鼠模型中随机选择8只作为糖尿病组(DM组),另8只DM大鼠经AD-MSC治疗(DM+MSCs组)。所有大鼠通过检测阴茎海绵体内压(ICP/MAP)等评估大鼠勃起功能,随后,取阴茎组织行组织学检查。结果 AD-MSCs在体内、外可分化为肌细胞和内皮细胞。AD-MSCs移植后,DM大鼠阴茎海绵体内压(ICP/MAP)值显著改善(P<0.05),阴茎组织中n NOS表达明显提高。结论 AD-MSCs可治疗糖尿病ED,其可能机制是通过提高阴茎海绵体n NOS阳性神经、内皮细胞的表达,这可能也与介导内源性MSCs聚集有关。 Objective To explore the possible mechanism of adipose-derived mesenchymal stem cells (AD-MSCs) transplantation to improve the relative erectile function of rats with diabetes mellitus (DM) associated erectile dysfunction (ED). Methods AD-MSCs were prepared, separated and developed to study their characteristics in vivo and in vitro. Newborn male rats were intraperitoneally injected with 5-ethynyl-2-deoxyuridine for tracking endogenous MSCs. 8 weeks later, eight of these rats were randomly selected as the normal control group (N group). The remaining rats were injected intraperitoneally with 60 mg/kg of streptozotocin (STZ) to induce DM. Eight of these DM rats were randomly selected as the DM group, while another eight rats were treated with AD-MSC (DM + MSCs group). All rats' erectile function were evaluated by intracavernous pressure (ICP/MAP) detection. Afterward, their penile tissues were collected for histological examination. Results AD-MSCs could differentiate into muscle cells and endothelial ceils in vivo and in vitro. After AD-MSCs transplantation, the ICP/MAP of DM rats significantly improved (P 〈0.05). The nNOS expression in penile tissues significantly increased. Conclusions AD-MSC can be used in the treatment of erectile function in rats with DM associated ED. The possible mechanism is to improve the expression of nNOS-positive nerves and endothelium in cavemosa, and may be related to mediate the recruitment of endogenous MSCs.
