四磨汤有效成分对慢性应激小鼠CCK及VIP的影响 被引量:1

Effect of effective constituents of Simo decoction on cholecystokinin andvasoactive intestinal peptide in mice with chronic stress

摘要 目的:探讨四磨汤有效成分木香烃内酯、槟榔碱、橙皮苷及乌药醚内酯对慢性应激小鼠胃窦CCK(胆囊收缩素)、VIP(血管活性肠肽)的影响。方法:将32只昆明种小鼠随机分为正常组、阴性对照组、四磨汤组及四磨汤成分组。采用饥饱失常、束缚夹尾等方法制备慢性应激小鼠模型,于应激第15天开始给予药物灌胃,连续7d。采用ELISA法测定胃窦组织匀浆CCK、VIP含量,IHC-P法测定胃窦组织CCK、VIP蛋白表达。结果:阴性对照组胃窦CCK、VIP含量及其蛋白表达均升高,与正常组比较差异有统计学意义(P<0.01);四磨汤成分组胃窦组织CCK、VIP含量及其蛋白表达均较阴性对照组降低,差异有统计学意义(P<0.05或P<0.01),而与四磨汤组比较差异无统计学意义(P>0.05)。结论:四磨汤发挥治疗功能性消化不良的作用机制可能与其有效成分木香烃内酯、槟榔碱、橙皮苷、乌药醚内酯降低胃窦组织中CCK、VIP水平有关。 Objective : To investigate the effect of effective constituents of Simo decoction ( costunolide, arecoline, hesperidin ,and linderane)on cholecystokinin(CCK) and vasoactive intestinal peptide(VIP) in the gastric antrum in mice with chronic stress. Methods:A total of 32 Kunming mice were randomly divided into normal group, negative control group, Simo decoction group,and Simo decoction constituent group. A nlouse model of chronic stress was established by irregular diet and tail clamp,and the drugs were given by gavage since day 15 of stress treatment for 7 consecutive days. ELISA was used to measure the content of CCK and VIP in the homogenate of the gastric antrum, and the IHC - P method was used to measure the expression of CCK and VIP in the gastric antrum. Results:Compared with the normal group,the negative control group had significant increases in the content and protein expression of CCK and VIP in the gastric antrum (P 〈0. 01 ). Compared with the negative control group, the Simo decoction constituent group had significant reductions in the content and protein expression of CCK and VIP in the gastric antrum(P 〈 0.05 or P 〈 0.01 ) , while there were no significant differences between the Simo decoction constituent group and the Simo decoction group ( P 〉 0.05 ). Conclusion : The mechanism of action of Simo decoction in the treatment of functional dyspepsia may be associated with the fact that its eft)ctive constituents, costunolide, arecoline,hesperidin,and linderane, can reduce the levels of CCK and VIP in the gastric antrum.
