附子多糖对小鼠海马糖原合成酶激酶-3β蛋白表达的影响 被引量:3

Effect of Radix Aconiti Lateralis Preparata polysaccharide on the protein expression of glycogen synthase kinase-3β in the hippocampus in mice

摘要 目的:探讨附子多糖对小鼠海马糖原合成酶激酶-3β(GSK-3β)蛋白表达的影响。方法:健康雄性C57BL/6J小鼠12只,分为对照组(n=3)和实验组(n=9),实验组分为附子多糖低、中、高3个剂量组,每组各3只,用药剂量分别为50mg/kg、100mg/kg、200mg/kg,对照组腹腔注射等量0.9%氯化钠注射液。6h后断头取脑,剥离海马组织,并用westren blot检测海马神经元中GSK-3β及p-GSK-3β的表达。结果:附子多糖各剂量组GSK-3β表达水平与对照组比较,差异无统计学意义(P>0.05),p-GSK-3β-Ser9/内参蛋白比值高于对照组(P<0.05)。结论:附子多糖影响GSK-3β活性可能是其增加神经细胞发生的作用机制之一。 Objective:To investigate the effect of Radix Aconiti Lateralis Preparata polysaccharide on the protein expression of glycogen synthase kinase- 36( GSK- 36)in the hippoeampus in mice. Methods:A total of 12 healthy male C57BL/6] mice were divided into control group with 3 mice and experimental group with 9 mice. The experimental group was further divided into low - , medium - , and high - dose Radix Aconiti Lateralis Preparata polysaeeharide groups, with 3 mice in each group and doses of 50mg/kg, 100mg/kg, and 200mg/kg, respectively. The mice in the control group were giv- en intraperitoneal injection of an equal volume of 0. 9% sodium chloride injection. The mice were decapitated and the brain was collected 6 hours later,the hippocampal tissue was peeled, and then Western blot was used to measure the expression of GSK - 3 β and p - GSK - 3 β in hippoeampal neurons. Results : There was no significant difference in the expression of GSK - 3β between the low - , medium - , and high - dose Radix Aconiti Lateralis Preparata polysaceharide groups and the control group (P 〉 0. 05 ) , and compared with the control group, the low - , medium - , and high - dose Ra- dix Aconiti Lateralis Preparata polysaecharide groups had a significantly higher p - GSK - 3β - Serg/internal reference protein ratio( P 〈 0. 05 ). Conclusion:Radix Aeoniti Lateralis Preparata polysaccharide may promote neuronal genesis by affecting the activity of GSK -3β.
