摘要
目的探讨砷对大鼠睾丸中肝细胞生长因子(hepatocyte growth factor,HGF)及其受体c-Met表达的影响。方法将40只健康清洁雄性SD大鼠随机分为4组,分别为对照(蒸馏水)组和2.4、12.0、60.0 mg/L亚砷酸钠染毒组,每组10只。采用自由饮用方式进行染毒,连续染毒6个月。测定大鼠睾丸组织内HGF及其受体c-Met的阳性表达及精子的凋亡情况。结果与对照组比较,各浓度亚砷酸钠染毒组大鼠睾丸组织内HGF、c-Met的阳性表达水平均较低,差异均有统计学意义(P<0.05);且随着亚砷酸钠染毒浓度的升高,大鼠的HGF、c-Met的阳性表达水平呈下降趋势。与对照组比较,各浓度亚砷酸钠染毒组大鼠的精子凋亡率均升高,差异有统计学意义(P<0.05);且随着亚砷酸钠染毒浓度的升高,大鼠的精子凋亡率呈上升趋势。结论慢性砷染毒可能通过抑制HGF-c-Met通路引起大鼠睾丸组织的损伤及精子凋亡。
Objective To explore the effects of arsenic on the expressions of hepatocyte growth factor(HGF) and its receptor c-Met in testis of rats. Methods Forty healthy clean grade male SD rats were randomly divided into four groups, respectively, the control group (distilled water),2.4, 12.0,60.0 mg/L sodium arsenite groups, 10 in each group. The rats were exposed to sodium arsenite by free drinking water for six consecutive months.The distribution of HGF and c-Met and apoptosis of spermatozoa in testis tissue were detected. Results Compared with the control group,the expression levels of HGF and c-Met in each exposure group were lower,the differences were statistically significant (P〈O.05);And with the increase of sodium arsenite concentrations, the positive expressions of HGF and c-Met in testis of rats were decreased. Compared with the control group,the apoptosis rates of spermatozoa in each sodium arsenite exposure group were higher, the differences were statistically significant (P〈0.05);And with the increase of sodium arsenite concentrations, the rates of sperm apoptosis were increased. Conclusion Chronic arsenic exposure can lead to the testis tissue injury and apoptosis by inhibiting the pathway of HGF-c- Met.
