摘要
目的研究水溶性维生素E(water-soluble vitamin E,WsVit E)对中波紫外线(ultraviolet-B,UVB)照射雄性小鼠生殖系统损伤的拮抗作用。方法将60只健康清洁级雄性昆明小鼠随机分6组,分别为空白对照组、WsVit E(30 mg/kg)对照组、辐射对照组及30、60、100 mg/kg WsVit E干预组,每组10只。WsVit E对照组及各剂量WsVit E干预组采用经口灌胃方式染毒WsVit E,空白对照组和辐射对照组灌胃生理盐水,染毒容量均为30 ml/kg;将辐射对照组及各剂量WsVit E干预组于UVB(285 nm)下辐照3 h,辐照强度为1.5 m W/cm^2;每天1次,连续处理30 d。测定小鼠精子活率及睾丸组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。结果与空白对照组相比,辐射对照组和各剂量WsVit E干预组小鼠的精子活率均较低,差异有统计学意义(P<0.05)。与辐射对照组相比,各剂量WsVit E干预组小鼠的精子活率均较高,差异有统计学意义(P<0.05);且随着WsVit E染毒量的升高,WsVit E干预组小鼠的精子活率呈先上升后下降的趋势,以60 mg/kg组为最高。辐射对照组和各剂量WsVit E干预组小鼠睾丸组织中的SOD活力均较低,而MDA含量均较高,差异有统计学意义(P<0.05)。与辐射对照组相比,各剂量WsVit E干预组小鼠睾丸组织中的SOD活力均较高,而MDA含量均较低,差异有统计学意义(P<0.05);且随着WsVit E染毒剂量的升高,WsVit E干预组小鼠睾丸组织中的SOD活力呈先上升后下降的趋势,而MDA含量呈先下降后上升的趋势,并均以60 mg/kg组为最高、最低值。结论 WsVit E对UVB辐射引起的雄性小鼠生殖系统损伤有一定的拮抗作用。
Objective To study the antagonism of water-soluble vitamin E(WsVitE) against the UVB-induced radiation damage to reproductive system in male mice. Methods Sixty Kunming male mice of clean grade were randomly divided into six groups, the blank control group,the WsVitE (30 mg/kg) control group,the radiation control group and 30, 60, 100 mg/kg WsVitE-treated groups,10 in each group. The WsVitE control group and the WsVitE-treated groups were given WsVitE, and the blank and radiation control groups were given normal saline by intragastric administration, the capacities were all 30 ml/kg. The radiation control group and WsVitE-treated groups were given three hours-UVB (285 nm) radiation once a day for 30 consecutive days, and the radiation intensity was 1.5 mW/cm2. The sperm motility rate,the SOD activity and the content of MDA of testicular tissue were determined. Results Compared with the blank control group, the sperm motility rate was lower in the radiation control group and Ws VitE-treated groups, with statistical significance (P〈0.05). Compared with the radiation control group, the sperm motility rate was higher in the WsVitE-treated groups with statistical significance (P〈0.05); And the sperm motility rates in the WsVitE-treated groups firstly increased and then decreased with the increase of WsVitE dosage, reached maximum in the 60 mg/kg WsVitE-treated group.Compared with the blank control group, the SOD activity was lower and MDA content was higher in the radiation control group and WsVitE-treated groups, with statistical significance (P〈0.05). Compared with the radiation control group, the SOD activity was higher, and MDA content was lower in the WsVitE-treated groups, with statistical significance (P〈0.05); And with the increase of WsVitE dosage, the SOD activity in the WsVitE-treated groups firstly increased and then decreased, while the MDA content firstly decreased and then increased, both the maximum SOD activity and minimum MDA content were in the 60 mg/kg WsVitE-treated group. Conclusion WsVitE has antagonism against the UVB-induced radiation damage to reproductive system in male mice to some extent.
