鹅源禽痘病毒的鉴定和基因分析 被引量:4

Identication and Phylogenetic Analyses of a Goose-originated Avipoxvirus

摘要 2016年6月,广西邕宁某场有20%的阳江鹅在喙、眼和脚出现赘肉状痘癍,临床无死亡病例。为了研究此病的病原和基因特征,采集痘癍样本、制备病理组织切片进行显微镜观察、并利用透射电镜进行形态学分析以及TaqMan荧光定量PCR检测,证实其病原为禽痘病毒属(Avipoxvirus,APV)成员,命名为YNE。通过对病毒核芯蛋白P4b编码基因(fpv167)和DNA聚合酶Popr编码基因(fpv094)的部分序列串联进行比对,发现该毒株与1991年分离于北美林鸳鸯的APV高度同源,属于A5.2基因分支。进一步序列分析发现,该毒株与同期发生在该场的麻鸭源APV相应序列的同源性可达99.9%,初步证实该病毒可以在鸭鹅间跨种传播。 We undertook pathogen and genetic characterization of a virus that caused an unidentified disease in Yangjiang geese in Yongning County (Guangxi Province, China) in June 2016. Clinical signs were cuta- neous nodules on the eyelids, beaks, and legs. About 20% of the Yangjiang geese were affected by this mystery disease but no deaths were observed. Clinical samples were collected and underwent histopatho- logic examination, electron microscopy analysis, and Taqman real-time polymerse chain reaction. An avi- poxvirus was identified as a pathogen of this disease outbreak, and designated "YNE". Furthermore, the partial sequences of the vial P4b gene (fpv167 locus) and DNA polymerase gene (fpv094 locus) were am- plified and analyses. The YNE group was in a new cluster within subclade A5.2, which comprises avipox- virus from an outbreak in wood duck in the USA in 1991. Nucleotide identity of 99.9% was also found be- tween YNE and YNY (which was isolated from mallard duck in the same farm). These data suggest that YNE could transmit from mallard duck to Yangjiang goose.
