摘要
目的探讨急性冠脉综合征患者血浆miR-15b、miR-92a的表达情况及其与冠脉病变严重程度的相关性方法收集138例研究对象,其中冠脉正常组44例,不稳定型心绞痛组(UA)44例,急性ST段抬高型心肌梗死组(STEMI)50例,应用RT-q PCR技术测定三组空腹血浆中miR-15b、miR-92a的表达水平,同时统计三组一般临床资料及生化指标。比较三组间生化指标、miRNAs水平的差异及miRNAs水平与冠脉SYNTAX评分的相关性。结果血浆miR-15b表达水平在STEMI组、UA组比冠脉正常组明显下降(P<0.01)。血浆miR-92a水平在STEMI组、UA组比冠脉正常组明显升高(P<0.01),同时,STEMI组miR-92a表达水平比UA组升高更显著(P<0.05)。STEMI组血浆miR-92a与SYNTAX评分呈正相关,相关系数r=0.779(P<0.01);UA组血浆miR-92a与SYNTAX评分呈正相关,相关系数r=0.828(P<0.01)结论 UA、STEMI患者血浆中miR-15b表达水平降低,而miR-92a表达水平升高,血浆miR-92a表达水平与急性冠脉综合征患者冠脉病变严重程度相关。
Objective To study the plasma expression levels of miR-15b, miR-92a in the patients with acute coronary syndrome and the correlation between the severity of coronary artery lesions. Methods we collected 138 patients and divided them into three groups,including 44 patients of normal coronary artery, 44 patients of unstable angina (UA), 50 patients of acute ST-elevated myocardial infarction (STEMI). The plasma levels of miR-15b and miR-92a were detected by RT-qPCR. Statistical software was used for statistical analysis to compare the difference between the three groups of basic information, biochemical indicators, the plasma levels of miRNAs, also the correlation between the levels of miRNAs with SYNTAX score. Results 1. The plasma levels of miR-15b in STEMI group and UA group were significantly decreased (P 〈 0.01 ) than normal coronary group, the differences were statistically significant. 2. The plasma levels of miR-92a in STEMI group and UA group were significantly increased (P 〈 0. 01 ) than normal coronary group, the differences were statistically significant; And the STEMI group of miR-92a levels was higher than the UA group (P 〈0.05) , the differences were statistically significant. 3. The plasma levels of miR-92a in STEMI group were positively correlated with the SYNTAX score, correlation coefficient r = 0. 779 ( P 〈 0.01 ) , The plasma levels of miR-92a in UA group were positively correlated with the SYNTAX score, correlation coefficient r =0. 828, (P 〈 0.01). Conclusion The plasma levels of miR- 15b decreased in patients with UA and STEMI, which could be used as a potential marker for the diagnosis of both. 2. The plasma levels of miR-92a increased in patients with UA and STEMI, which could be used as a potential marker for the diagnosis of both. 3. The plasma levels of miR-92a could be used as a potential marker for assessing the severity of coronary artery disease in patients with ACS.
