摘要
以野生中华散尾鬼笔为研究对象,对其菌丝进行分离纯化,以菌丝的生长速率为指标,对中华散尾鬼笔的培养基及培养条件进行优化,筛选出适宜中华散尾鬼笔菌丝生长的基础培养基,优化碳源、氮源、无机盐添加量及pH、培养温度等培养条件。结果表明,中华散尾鬼笔适宜培养基配方(每L)为:马铃薯200g、葡萄糖20g、蛋白胨3g、琼脂15g、10%无机盐溶液(KH_2PO_4 1g、K_2HPO_41g、Mg_SO_40.5g、FeSO_40.01g、NaNO_30.5g、CaCl_20.05g、KCl 0.5g溶于100mL蒸馏水);最佳培养条件pH为5.0、温度22℃,在此培养条件下,中华散尾鬼笔菌丝的净生长速率为0.59cm·d^(-1),与培养基A相比增长23.69%。
Lysurus mokusin extract has insecticidal and antifungal activities,and it is necessary to cultivate a large number of mycelium because its mycelia grow slowly in the laboratory environment. In order to im- prove the growth rate of Lysurus rnokusin mycelia, the culture medium and conditions were screened and optimized in this study. The culture medium was screened with the growth rate of mycelium as the evalua- tion index,and the suitable medium for the growth of L. mokusin mycelia was selected out. And on this ba- sis, the carbon source, nitrogen source, inorganic salt addition, pH and culture temperature were further op- timized. The best culture medium was 200 g of potato,20 g of glucose,3 g of peptone, 10% inorganic salt solution (KH2PO4 1 g,K2HPO4 1 g,MgSO4 0.5 g,CaCl2 0.05 g,KCl 0.5 g,FeSO4 0.01 g and NaNO3 0.5 g dissolved in 100 mL distilled water) and 15 g of agar. The optimum culture conditions were pH= 5 and temperature of 20℃. Under the above culture conditions, the net growth rate of L. mokusin mycelia was 0. 59 cm/d,which was 23.69% higher than that of medium A.
