摘要
目的:观察参芪扶正注射液对免疫低下小鼠免疫功能的调节作用以及核因子-kappaB(NF-κB)和IκBα蛋白表达的影响。方法:将48只C57BL/6小鼠随机分为空白对照组、模型组、参芪扶正注射液低剂量组和高剂量组,每组12只。除空白对照组外,各组小鼠给予环孢素A诱导建立免疫低下模型,同时参芪扶正注射液低、高剂量组分别每天腹腔注射参芪扶正注射液15 mL/kg、30 mL/kg,连续给药14 d。第15天小鼠禁食麻醉后,取血离心取血清,采用ELISA法检测各组小鼠血清中白细胞介素-2(IL-2)和γ-干扰素(IFN-γ)水平。取脾脏称重计算脾脏指数,检测脾脏中IL-2、IFN-γ的表达,以及脾脏组织中NF-κB和IκBα蛋白的表达。结果:与模型组比较,参芪扶正注射液低、高剂量组脾脏指数有显著的升高(P<0.05);与模型组比较,血清、脾脏中的IL-2、IFN-γ在参芪扶正注射液低、高剂量组中显著升高(P<0.05);与模型组比较,参芪扶正注射液低、高剂量组脾脏细胞核内的NF-κB蛋白表达显著升高;细胞质内的IκBα蛋白表达明显降低。结论:在免疫低下小鼠模型中,参芪扶正注射液能够通过提高脾脏指数、升高IL-2、IFN-γ的表达,调节NF-κB、IκBα蛋白的表达,以达到提高机体免疫功能的目的。
Objective: To investigate the effect of Shenqi Fuzheng Injection(参芪扶正注射液,SQFZI) on immunological function and expression of NF-κB,IκBα in immunosuppressed mice mode.Methods: Forty-eight C57 BL/6 mice were divided randomly into the four groups,including control group,model group,low dose(15 mL/kg) of SQFZI group,and high dose(30 mL/kg) of SQFZI group.All mice were made into immunosuppressed models induced by Cs A injection,except that in control group.Low dose and high dose groups were treated with SQFZI at a dose of 15 mL/kg or 30 mL/kg.All mice were killed after 14 days.Spleens were removed and calculated spleen index.Expression of IL-2 and IFN-γ in serum or spleen cells were detected using ELISA kit.Western blot was used to assay protein expression of NF-κB,IκBα in spleens.Results: The spleen index of mice in SQFZI low and high dose groups were significantly higher than model group(P〈0.05).Compared with model group,IL-2 and IFN-γ expression of SQFZI low and high dose groups increased significantly(P〈0.05).The protein expression of NF-κB in the nucleus increased,and the expression of IκBα decreased in the cytoplasm,compared with model group.Conclusion: SQFZI could enhance immunological function by effecting spleen index,expression of IL-2,IFN-γin serum or spleen cells,and regulating NF-κB,IκBαproteins.
